Description: Caspian trout (Salmo trutta caspius) is one of valuable fishes of the Caspian sea for people living in the Northern coastal area of Iran. Since fish stock and natural resources protection is one the Iranian fisheries research objectives, the most activity on Caspian trout rehabilitation was focused on releasing thousands of smolts in the rivers discharge to Caspian Sea. Catch statistics of this species has declined over last decades thus this study on quality of provided brood stocks and producing fries with suitable quality can help rehabilitation and rearing of this valuable species. Several rivers were considered for providing brooders to keep genetic resources variety of sea stocks. In spawning season 25 (9 male, 16 female) and 30 (5 male, 25 female) brooders were collected from fishermen in Mazandaran rivers and Gyilan coasts provinces respectively. After propagation and larvae rearing, 11500 (10-15 g.) and 50000 (less than 10 g.) juveniles were released in Gyilan and Mazandaran rivers respectively.

Description: In this study the genetic diversity of wild Caspian trout (Salmo trutta caspius) in the Sardabroud and Astara Rivers was evaluated using D- Loop region sequencing. For this purpose, 35 specimens of adult Caspian brown trout were collected from these rivers in the Mazandarn and Gilan Provinces in fall and winter 2011. Approximately 3-5 g of soft and fresh fin tissue was isolated and fixed in ethanol 96% and then transferred to the Caspian Sea Ecology Research Center Genetics Laboratory in Sari, Iran. Genomic DNA from the samples was extracted using Ammonium Acetate Method. The quality and quantity of the extracted DNA were assessed by spectrophotometer and agarose gel (1%) electrophoresis. Polymerase Chain Reaction (PCR) was performed on the target DNA using a primers sequence D- Loop region of mtDNA molecule. Then the product was purified and DNA sequencing was carried out using chain termination method. The D- Loop region of Caspian trout contained 654 bp. Data were analyzed using Bio-Edit, DnaSP, Arlequin and Mega software. 20 and 15 haplotypes was observed in Sardabrud and Astara River. Tthe DNA sequence of one of them was recorded in Gene Bank with numbers KC991027 and KF015727. 223 and 240 polymorphic loci were detected in Sardabrud and Astara River that all of them were out of Hardy- Weinberg equilibrium (p〈0.05). Average nucleotide and haplotype diversity were 0.127±0.067, 1.000±0.005 in Sardabrud River and 0.118±0.063 and 1.000±0.005 in Astara River.

Description: The fishing industry is deeply dependent on complex and high quality diets consisting of dry feed, vitamins and micro/macro-elements. The suitable dose of each essential nutrient is different based on the fish species, their age and physiological conditions. In this study, the effects of two recommended doses of riboflavin (20 and 30 mg vitamin per each kg dry feed) on growth, hematological and immunological parameters of rainbow trout (Oncorhynchus mykiss) fingerlings were investigated. The results showed that although treatment with riboflavin could improve biometric and immunological parameters of the fishes but the higher concentration (30mg) of riboflavin had no extra-positive effects on these factors. Furthermore, both doses decreased hematopoietic activity of this fish. Overall, the results of this study showed that treatment with 20 mg of riboflavin is more effective than treatment with 30 mg of riboflavin per kg of dried food.

Description: This study was conducted to evaluate the effects of replacing dietary fish oil with canola oil on growth indices, survival rate and body chemical composition of rainbow trout (Oncorhynchus mykiss) fingerling. For this purpose, three experimental diets were prepared with the same protein level (40%) and fat levels of 100% )First diet with 10% fish oil, second diet with 100% canola oil and third diet with 50% fish oil and 50% canola oil with three replicates). Then 405 fingerling rainbow trout with an average weight of 11±1 g and a density of 45 in 9 ponds were fed in experimental diets for 8 weeks. Based on the results, the combination of 5% fish oil and 5% canola oil in the ration improved the growth indices including: final weight, Increase Body Weight (BWI), Special Growth Ratio (SGR) and Condition Factor (CF) and showed a significant difference with other treatments and the highest amount was observed in 10% fish oil treatment (p〈0.05). However, there was no significant difference between treatments in terms of survival rate and protein, fat, ash and moisture content of fish carcasses (p〉0.05). Based on the results of this study, replacing part of canola oil with fish oil (50% fish oil with 50% canola oil) in rainbow trout diet has a positive effect on growth indices and can reduce costs by reducing the length of breeding period.
