Notes: Abstract Cinnamomin is a plant type II ribosome-inactivating protein (RIP) isolated from the seeds of Cinnamomum camphora. It consists of two nonidentical polypeptide chains (A- and B-chain) held together through one disulfide linkage. Its A- and B-chain contain 0.3% and 3.9% sugars respectively. The B-chain of cinnamomin was digested by pronase E and then the liberated glycopeptides were separated from non-glycopeptides by gel filtration chromatography on a Bio-Gel P-4 column. Three crude glycopeptides were obtained by continuing chromatography over anion-exchange resin (AG1-X2) in the buffer of 2% pyridine-acetic acid (pH 8.3) with a polygradient elution system. Through further purification by the gel filtration chromatography and HPLC, three major glycopeptides, GP1, GP2 and GP3 were obtained. Mainly by two-dimensional Nuclear Magnetic Resonance (NMR) including TOCSY, DQF-COSY, NOESY, HMQC and HMBC, their primary structures were analyzed as: Manα1,3Manα1,6(Manα1,3)(Xylβ1,2)Manβ1,4GlcNAcβ1,4GlcNAcβ1-(Gly-)Asn-Asn-Thr(GP1), Manα1,6(Manα1,3)(Xylβ1,2)Manβ1,4GlcNAcβ1,4(Fucα1,3)GlcNAcβ1-Asn-Ala-Thr(GP2),Manα1,6(Manα1,3)Manα1,6(Manα1,2 Manα1,3)Manβ1,4GlcNAcβ1,4GlcNAcβ1-(Ala-)Asn-Gly-Thr(GP3).

Notes: Glycolysis of poly(ethylene terephthalate) with bisphenol A at high temperature (190°C-230°C) was carried out in an autoclave reactor. Kinetic study on depolymerization of glycolysis was conducted. The reaction temperature and the ratio of bisphenol A to poly(ethylene terephthalate) played the key roles on the reaction rate and the extent of glycolysis. For describing the kinetic behavior properly, a mechanism consisting of six principle equilibrium reactions was proposed. Based on this mechanism, the kinetic parameters were obtained through the Runge-Kutta numerical integration. Excellent agreements between the calculated values and the experimental data were achieved, thus verified the appropriateness of the proposed mechanism.

Notes: Cinnamomin (two-chain) and camphorin (single-chain), two novel ribosome-inactivating proteins (RIPs) purified from the seeds of Cinnamomum camphora, produced inhibitory effects in cultured carcinoma cells. The IC50 of cinnamomin to the human hepatocarcinoma cell-line 7721 and the melanoma cell-line M21 were 18·8 nmol and 11·7 nmol respectively. The IC50 of camphorin to the human hepatocarcinoma cell-line 7721 was 59 nmol, whereas the melanoma cell-line M21 was not susceptible to camphorin. Furthermore, cinnamomin exhibited a remarkable inhibitory effect on the growth of solid melanoma in the skin of the nude mouse. An R-fragment could be isolated from ribosomes of cinnamomin- or camphorin-treated carcinoma cells after incubation with acidic aniline, indicating that the cytotoxicity of these two new RIPs to carcinoma cells might result from modification to the ribosomes.

Description: Pyrin, encoded by theMEFVgene, is best known for its gain-of-function mutations causing familial Mediterranean fever (FMF), an autoinflammatory disease. Pyrin forms a caspase-1–activating inflammasome in response to inactivating modifications of Rho GTPases by various bacterial toxins or effectors. Pyrin-mediated innate immunity is unique in that it senses bacterial virulence rather than microbial molecules, but its mechanism of activation is unknown. Here we show that Pyrin was phosphorylated in bone marrow-derived macrophages and dendritic cells. We identified Ser-205 and Ser-241 in mouse Pyrin whose phosphorylation resulted in inhibitory binding by cellular 14-3-3 proteins. The two serines underwent dephosphorylation upon toxin stimulation or bacterial infection, triggering 14-3-3 dissociation, which correlated with Pyrin inflammasome activation. We developed antibodies specific for phosphorylated Ser-205 and Ser-241, which confirmed the stimuli-induced dephosphorylation of endogenous Pyrin. Mutational analyses indicated that both phosphorylation and signal-induced dephosphorylation of Ser-205/241 are important for Pyrin activation. Moreover, microtubule drugs, including colchicine, commonly used to treat FMF, effectively blocked activation of the Pyrin inflammasome. These drugs did not affect Pyrin dephosphorylation and 14-3-3 dissociation but inhibited Pyrin-mediated apoptosis-associated Speck-like protein containing CARD (ASC) aggregation. Our study reveals that site-specific (de)phosphorylation and microtubule dynamics critically control Pyrin inflammasome activation, illustrating a fine and complex mechanism in cytosolic immunity.

Description: Based on the data of green credit (GC), environmental regulation (ER) and green technology innovation (GTI) in 30 provinces and cities of China from 2007 to 2016, this study investigated the relationship between green credit and green technology innovation development and analyzed the adjustment effect of ER on GC to promote GTI using Geoda and Matlab2016 software, so as to further guide and encourage GC. The results show that GTI in 30 provinces and municipalities in China has a significant spatial agglomeration effect. Single GC plays a certain role in promoting local technology innovation, but it fails to influences the surrounding areas. Environmental regulation has a certain regulatory effect on the relationship between green credit and green technology innovation in the province but also fails to influences the surrounding areas.

Description: In this study, Multiwalled carbon nanotubes (MWCNTs) were oxidized by a mixture of sulfuric acid and nitric acid (V:V = 3:1) at 70 °C for 1, 2, and 4 h, respectively. The oxidized MWCNTs were characterized by N2 adsorption, Fourier-transform infrared spectroscopy (FT-IR), X-ray photoelectron spectroscopy (XPS), thermal gravimetric analysis (TGA), and Raman spectroscopy to determine the oxidation degree. The dispersion of the MWCNTs was investigated by UV-vis-NIR, SEM, and dynamic light scattering measurements. Results show that sulfonitric treatment increased the surface area and total pore volume and reduced the average pore diameter of MWCNTs. The treatment promoted the formation of oxidized species on the surface MWCNTs, as identified by FT-IR, TGA, and X-ray photoelectron spectroscopy measurements, and more oxygen-containing functional groups were generated when treatment time was extended. Moreover, a general relationship between oxidation degree and dispersibility of MWCNTs in water was established. UV-vis-NIR and dynamic light scattering measurements and SEM images revealed that MWCNTs with higher oxidation degree showed better dispersibility in water.

Description: Firms’ performance in sustainable development has caused increasing concerns. A key issue is that the environmental characteristics of business managers may be an important driver to promote the sustainable practice of enterprises. This study explores the relationship between ascribed responsibility of managers and green sustainable practice of enterprises by introducing the waste management as mediation variable and green self-efficacy as a moderating variable. A survey was conducted in manufacturing enterprises from the Yangtze River Delta in China in 2019, where 149 valid questionnaires were collected out of 200 surveyed enterprises. Subsequently, the data were analyzed through an analytic hierarchy process. The results show that: (1) ascribed responsibility of managers has a positive impact on green sustainable practices (β = 0.428, p 〈 0.001), and waste management plays a full mediating role during this process (β = 0.428, p 〈 0.001); (2) managers' green self-efficacy strengthens the positive impact of waste management on sustainable practices (β = 0.284, p 〈 0.05); (3) green self-efficacy has positively moderated the indirect effect of managers' ascribed responsibility for sustainable practice through waste management. The research enriches the theories on sustainable practice of enterprises from the perspective of managers’ environmental characteristics and provides insights for enterprises to promote sustainable development practice.

Description: This paper proposes a strain prediction method for wind turbine blades using genetic algorithm back propagation neural networks (GA-BPNNs) with applied loads, loading positions, and displacement as inputs, and the study can be used to provide more data for the wind turbine blades’ health assessment and life prediction. Among all parameters to be tested in full-scale static testing of wind turbine blades, strain is very important. The correlation between the blade strain and the applied loads, loading position, displacement, etc., is non-linear, and the number of input variables is too much, thus the calculation and prediction of the blade strain are very complex and difficult. Moreover, the number of measuring points on the blade is limited, so the full-scale blade static test cannot usually provide enough data and information for the improvement of the blade design. As a result of these concerns, this paper studies strain prediction methods for full-scale blade static testing by introducing GA-BPNN. The accuracy and usability of the GA-BPNN prediction model was verified by the comparison with BPNN model and the FEA results. The results show that BPNN can be effectively used to predict the strain of unmeasured points of wind turbine blades.
