    Notes: We tested if genetic exchange was observable between two strains of Leishmania major (Trypanosomatidae) during mixed infection of the sand fly Phlebotomus papatasi. Previous studies suggested that genetic exchange may occur in natural populations of Leishmania at a low frequency, but experimental crosses examining small numbers of progeny (〈60) did not reveal hybrid parasites. Accordingly, a strategy was devised to increase the number of progeny that could be screened by 100-fold. Clonal derivatives from two strains that were infective to flies and contained numerous restriction fragment length polymorphisms were characterized and selected for resistance to methotrexate or tunicamycin by gene amplification. A successfully mixed infection of P. papatasi was obtained, and a method was developed for directly plating promastigotes from the gut contents of infected flies onto selective media. Twenty-five hundred independent progeny were scored for the presence of both drug resistance markers. No hybrid parasites were observed, indicating that the frequency of genetic exchange in this cross must be less than 4 times 10-4. The lines and methods established in this work may prove useful in future studies of the mechanism and frequency of gene exchange in Leishmania.

    Notes: The potential energy curve of the ground electronic state of the Hg dimer has been calculated using the CCSD(T) procedure and relativistic effective core potentials. The calculated binding energy (0.047 eV) and equilibrium separation (3.72 Å) are in excellent agreement with experiment. A variety of properties, including the second virial coefficient, rotational and vibrational spectroscopic constants, and vibrational energy levels, have been calculated using this interatomic potential and agreement with experiment is good overall. © 2001 American Institute of Physics.

    Notes: The one- and two-photon properties of free base porphin, free base porphin dianion, and the 2,4-substituted diformyl and divinyl analogs of these molecules are studied using a semiempirical SCF-MO formalism (CNDO-π-SCF-MO-PSDCI) including extensive single and double configuration interaction. Strongly two-photon allowed states are predicted to lie in the Soret region as well as in the region between the Soret and visible bands. A number of the two-photon allowed states in the Soret region are predicted to have two-photon absorptivities exceeding 100×10−50 cm4 s molecule−1 photon−1. The calculations indicate that the visible (Q) states are well characterized by the four orbital model, whereas the Soret (B) states contain significant contributions from configurations comprised of other orbitals. The inclusion of extensive double configuration interaction significantly reduces the Soret-visible (B–Q) splitting, increases the Qx–Qy splitting, and yields calculated oscillator strengths for the Q bands in better agreement with experiment than values calculated using single CI alone. The effects of conjugation into the porphyrin macrocycle are predicted to be more significant than inductive effects on macrocycle π orbitals due to substituent polarity. The 〈Qx||r||S0〉 and 〈Qy||r||S0〉 transition moment vectors are predicted to lie approximately through adjacent pyrrole rings in 2- and 4-monoformyl free base porphin dianions and approximately through adjacent methine bridges in 2,4-diformyl free base porphin dianion.

    Notes: Inter-generational effects on the growth of Poa annua (L.) in ambient and elevated atmospheric CO2 conditions (350 and 550 μl l–1, respectively) were studied in two different experiments. Both experiments showed similar results. In a greenhouse experiment growth, measured as the numbers of tillers produced per week, was compared for plants grown from first and second generation seeds. Second generation seeds were obtained from plants grown for one whole generation in either ambient or elevated atmospheric CO2 (‘ambient’ and ‘elevated’ seeds, respectively). First generation plants and second generation ‘ambient’ plants did not respond to elevated CO2. Second generation ‘elevated’ plants produced significantly more tillers in elevated CO2. In the second experiment model terrestrial ecosystems growing in the Ecotron and which included Poa annua were used. Above-ground biomass after one and two generations of growth were compared. At the end of Generation 1 no difference was found in biomass production while at the end of Generation 2 biomass increased in elevated CO2 by 50%. The implications for climate change research are discussed.

    Notes: Although soil organisms play an essential role in the cycling of elements in terrestrial ecosystems, little is known of the impact of increasing atmospheric CO2 concentrations on soil microbial processes. We determined microbial biomass and activity in the soil of multitrophic model ecosystems housed in the Ecotron (NERC Centre for Population Biology, Ascot, UK) under two atmospheric CO2 concentrations (ambient vs. ambient + 200 ppm). The model communities consist of four annual plant species which naturally co-occur in weedy fields and disturbed ground throughout southern England, together with their herbivores, parasitoids and soil biota. At the end of two experimental runs lasting 9 and 4.5 months, respectively, root dry weight and quality showed contradictory responses to elevated CO2 concentrations, probably as a consequence of the different time-periods (and hence number of plant generations) in the two experiments. Despite significant root responses no differences in microbial biomass could be detected. Effects of CO2 concentration on microbial activity were also negligible. Specific enzymes (protease and xylanase) showed a significant decrease in activity in one of the experimental runs. This could be related to the higher C:N ratio of root tissue. We compare the results with data from the literature and conclude that the response of complex communities cannot be predicted on the basis of oversimplified experimental set-ups.

    Notes: Restriction and modification (R–M) systems are generally thought to protect bacteria from invasion by foreign DNA. This paper proposes the existence of an alternative role for the phase-variable R–M systems encoded by the hsd loci of Mycoplasma pulmonis. Populations of M. pulmonis cells that arose during growth in different environments were compared with respect to R–M activity and surface antigen production. When M. pulmonis strain X1048 was propagated in laboratory culture medium, 〉 95% of colony-forming units (cfu) lacked R–M activity and produced the variable surface protein VsaA. Mycoplasmas isolated from the nose of experimentally infected rats also lacked R–M activity and produced VsaA. In contrast, the cell population of mycoplasmas isolated from the lower respiratory tract of the infected rats was more complex. The most dramatic results were obtained for mycoplasmas isolated from the trachea. At 14 days postinfection, 38% of mycoplasma isolates produced a Vsa protein other than VsaA, and 34% of isolates had active restriction systems. These data suggest that differences in selection pressures in animal tissues affect the surface proteins and the R–M activity of the mycoplasmal cell population. We propose that variations in the production of R–M activity and cell surface proteins are important for the survival of the mycoplasma within the host.
