    Notizen: Abstract The solubility properties and the number of disulfide groups per molecule of the crystal protein of Bacillus thuringiensis were shown to be similar to that of wool keratin. Dissolution of the crystals required scission of S-S bonds. This could be achieved at neutral pH without loss of toxicity. Molecular weight determinations with gel electrophoresis and ultracentrifugation indicated that the crystal subunit is a dimer. With the exception of the variety israelensis, all strains belonging to ten different subspecies revealed crystal subunits of the same molecular weight.

    Notizen: Abstract In addition to the previously described gene yptV1 encoding a small G-protein we have now identified and sequenced four more ras-related ypt genes (yptV2-yptV5) from the green alga Volvox carteri. The four new genes encode polypeptides consisting of 203 to 217 amino-acid residues that contain the typical sequence elements (GTP-binding domains, effector domain) of the ypt/rab subgroup of the Ras superfamily. Comparison of the derived amino-acid sequences from the V. carteri ypt gene products and their Ypt homologs from other species revealed similarity values ranging from 60% to 85%, whereas intraspecies similarities were found to approach only 55%. The coding sequences are interrupted by 5–7 introns of variable size (70–1000 nucleotides) occupying different positions in the genes. Reverse-transcribed samples of stage-specific RNAs were PCR-amplified with primers specific to yptV1, yptV3, yptV4, and yptV5 to determine if yptV transcription might be restricted to either cell type or to a specific stage of the life cycle. These experiments demonstrated that each of these genes is expressed throughout the entire Volvox life cycle and in both the somatic and the reproductive cells of the alga. The transcription start sites of yptV1 and yptV5 were mapped by primer extension. Expression of recombinant yptV cDNA in E. coli yielded recombinant proteins that bound GTP specifically, demonstrating a property which is typical for small G-proteins. The derived YptV polypeptide sequences were used to group them into four distinct classes of Ras-like proteins. These are the first proteins of the Ras superfamily to be identified in a green alga. We discuss the possible role of the YptV-proteins in the intracellular vesicle transport of Volvox.

    Notizen: Zusammenfassung 1. Die B · I aus der Kreuzung Oe.Berteriana (B · l) × odorata (v · I) hat in der Diakinese der Meiosis die Chromosomenanordnung 6+6+2. 2. Obwohl demzufolge bei Selbstungen und Kreuzungen Spaltungen zu erwarten wären, konnten doch immer nur die B- und I-Komplexe, nie aber Austauschkomplexe nachgewiesen werden. 3. Durch anatomische Untersuchungen des Fruchtknoteninhaltes, durch Auszählen von Kreuzungskapseln sowie Bestimmen der Pollenfertilität wurde festgestellt, daß der Anteil an sterilen Samenanlagen und Pollenkörnern bei der B · I viel höher ist als bei der B · l. 4. Diese Sterilitätserhöhung bei der B · I ist sicher durch das aus unbekannten Gründen erfolgende Absterben der Austauschgonen bedingt. 5. Ob bei der B · I die Austauschgonen so häufig sind wie die mit dem B-bzw. I-Komplex, dabei die Grundsterilität nur 14% gegenüber den 40% bei der B · l beträgt, oder ob bei verminderter, jedoch nicht bestimmbarer Grundsterilität infolge Polarisation mehr Gonen mit dem B- und I-Komplex als hernach absterbende Gonen mit den Austauschkomplexen gebildet werden, kann nicht entschieden werden.

    Notizen: Abstract The Ypt/Rab proteins are small GTPases, which belong to the Ras superfamily and have been shown to be involved in endo-and exocytosis in mammalian cells and yeast. Using affinity-purified antibodies specific for four Ypt proteins, namely Ypt1p, Ypt4p, Ypt5p and Ypt6p, of the multicellular green alga Volvox carteri (YptVp) and its close unicellular relative Chlamydomonas reinhardtii (YptCp), we examined the abundance of the corresponding antigens during the asexual life cycle of Volvox, and their intracellular localization. The YptV proteins were found in all stages throughout the asexual life cycle and are tightly associated with intracellular membranes. Indirect immunofluorescence revealed that YptV4p, YptV5p and YptV6p are present in perinuclear regions of the cell, indicating an association with the Golgi region. Golgi localization of YptV4p and YptV6p in Volvox was confirmed by immunogold electron microscopy. In contrast, we found Ypt1p associated with the contractile vacuole in both V. carteri and C. reinhardtii. Furthermore, the YptV proteins were also detected along the entire length of the flagella of somatic Volvox cells. This flagellar location was substantiated by western blot analysis of extracts prepared from isolated flagella of both algae. While localization to exocytic compartments is in agreement with the established Ypt/Rab function in intracellular vesicle transport of eukaryotic cells, presence in the algal flagellum is the first hint of a possible role for small G proteins also in motility organelles.

    Notizen: The adsorption of several toluene-soluble polymers at the toluene-water interface has been investigated by using the duNouy ring method of measuring interfacial tension γT/W. Polystyrene and poly(ethylene-co-vinyl acetate) (11.1 mole-% vinyl acetate) have little affinity for this interface at 29°C, but poly(methyl methacrylate) (PMMA) (M̄n = 420,000) and ethyl cellulose (EC) (M̄n = 50,100; 49.1% ethoxyl) adsorb significantly at concentrations as low as 1.0 × 10-4 g/100ml. A plot of interfacial tension lowering versus initial logarithm of initial bulk phase polymer concentration is linear from 1.0 × 10-4 to 1.0 × 10-1 g/100 ml for EC and 1.0 × 10-4 to 1.0 × 10-2 g/100 ml for PMMA. When the PMMA concentration increases to 1.15 × 10-1 g/100 ml, its adsorption behavior changes markedly. Prolonged time effects occur and adsorption becomes dependent upon dissolved water content of the toluene prior to formation of the toluene/water interface. Such effects are not observed with the other solutions studied. Increasing temperatures have variable effects on values of γT/W for the polymer solutions studied. Experiments with various polymer mixtures indicate that the polymer lowering T/W the most is preferentially adsorbed at the toluene-water interface and rapidly displaces less strongly adsorbed polymers.
