    Description: A variant form of a group I ribozyme, optimized by in vitro evolution for its ability to catalyze magnesium-dependent phosphoester transfer reactions involving DNA substrates, also catalyzes the cleavage of an unactivated alkyl amide when that linkage is presented in the context of an oligodeoxynucleotide analog. Substrates containing an amide bond that joins either two DNA oligos, or a DNA oligo and a short peptide, are cleaved in a magnesium-dependent fashion to generate the expected products. The first-order rate constant, kcat, is 0.1 x 10(-5) min-1 to 1 x 10(-5) min-1 for the DNA-flanked substrates, which corresponds to a rate acceleration of more than 10(3) as compared with the uncatalyzed reaction.

    Description: The spray characteristics of a spill-return airblast atomizer are examined using water as the working fluid. Measurements of mean drop size, drop size distribution, spray cone angle, and circumferential liquid distinction, are carried out over wide ranges of liquid injection pressures and atomizing air velocities. Generally, it is found that an increase in nozzle bypass ratio worsens the atomization quality and widens the spray cone angle. Increase in airblast air velocity may improve or impair atomization quality depending on whether it increases or decreases the relative velocity between the liquid and the surrounding air. Airblast air can also be used to modify the change in spray cone angle that normally accompanies a change in bypass ratio.

    Description: The Tetrahymena group I ribozyme was modified by replacing all 99 component uridine residues with 5-bromouridine. This resulted in a 13-fold reduction in catalytic efficiency in the RNA-catalyzed phosphoester-transfer reaction compared to the behavior of the unmodified ribozyme. A population of 10(13) variant ribozymes was constructed, each containing 5-bromouridine in place of uridine. Five successive 'generations' of in vitro evolution were carried out, selecting for improved phosphoester transferase activity. The evolved molecules exhibited a 27-fold increase in catalytic efficiency compared to the wild-type bromouridine-containing ribozyme, even exceeding that of the wild-type ribozyme in the non-brominated form. Three specific mutations were found to be responsible for this altered behavior. These mutations enhanced activity in the context of 5-bromouridine, but were detrimental in the context of unmodified uridine. The evolved RNAs not only tolerated but came to exploit the presence of the nucleotide analogue in carrying out their catalytic function.

    Description: We obtained 98 R-band and 18 B, r', i' images of the optical afterglow of GRB 060526 (z = 3.21) with the MDM 1.3 m, 2.4 m, and the PROMPT telescopes at CTIO over the five nights following the burst trigger. Combining these data with other optical observations reported in GCN and the Swift XRT observations, we compare the optical and X-ray afterglow light curves of GRB 060526. Both the optical and X-ray afterglow light curves show rich features, such as flares and breaks. The densely sampled optical observations provide very good coverage at T 〉 10(exp 4) s. We observed a break at 2.4 x 10(exp 5) sin the optical afterglow light curve. Compared with the X-ray afterglow light curve, the break is consistent with an achromatic break supporting the beaming models of GRBs. However, the prebreak and postbreak temporal decay slopes are difficult to explain in simple afterglow models. We estimated a jet angle of theta(sub j) approx. 7deg and a prompt emission size of R(sub prompt) approx. 2 x 10(exp 14) cm. In addition, we detected several optical flares with amplitudes of (Delta)m approx. 0.2,0.6, and 0.2 mag. The X-ray afterglows detected by Swift have shown complicated decay patterns. Recently, many well-sampled optical afterglows also show decays with flares and multiple breaks. GRB 060526 provides an additional case of such a complex, well-observed optical afterglow. The accumulated well-sampled afterglows indicate that most of the optical afterglows are complex.

    Description: Microgravity significantly reduces transmission of ground-reaction forces to bones, promoting atrophy. There is little information available concerning the effects of microgravity on bones at sites where anti-gravity muscles are attached (tendon-bone junctions). This study evaluates the effects of microgravity on the origin and insertion sites of anti-gravity muscles on the rat tibia, fibula and calcaneus. Changes in the strength of those tendon-bone junctions could predispose the animal to injury following spaceflight.

    Description: Stochastic differential equations (SDEs) are mathematical models that are widely used to describe complex processes or phenomena perturbed by random noise from different sources. The identification of SDEs governing a system is often a challenge because of the inherent strong stochasticity of data and the complexity of the system’s dynamics. The practical utility of existing parametric approaches for identifying SDEs is usually limited by insufficient data resources. This study presents a novel framework for identifying SDEs by leveraging the sparse Bayesian learning (SBL) technique to search for a parsimonious, yet physically necessary representation from the space of candidate basis functions. More importantly, we use the analytical tractability of SBL to develop an efficient way to formulate the linear regression problem for the discovery of SDEs that requires considerably less time-series data. The effectiveness of the proposed framework is demonstrated using real data on stock and oil prices, bearing variation, and wind speed, as well as simulated data on well-known stochastic dynamical systems, including the generalized Wiener process and Langevin equation. This framework aims to assist specialists in extracting stochastic mathematical models from random phenomena in the natural sciences, economics, and engineering fields for analysis, prediction, and decision making.
