    Notes: The de novo purine biosynthetic enzymes 5-amino-4-imidazolecarboxamide-ribonucleotide (AICAR) transformylase (EC 2.1.2.3), IMP cyclohydrolase (EC 3.5.4.10) and glycineamide-ribonucleotide (GAR) synthetase (EC 2.1.2.2) are encoded by the purHD locus of Escherichia coli. The DNA sequence of this locus revealed two open reading frames encoding polypeptides of Mr57335 and 45945 (GAR synthetase), respectively, that formed an operon. The DNA sequence, maxicell and complementation analyses all supported the concept that the Mr 57335 polypeptide is the product of the purH gene and encodes a bifunctional protein containing both AICAR trans-formylase and IMP cyclohydrolase activities. The 5′ end of the purHD mRNA was determined by primer extension mapping and contains two regions of dyad symmetry capable of forming ‘hairpin’ loops where the formation of the one would prevent the formation of the other but not vice versa. Regulation by the purR gene product was explained by the discovery of a purR binding site in the purHD control region.

    Notes: Aspergillus fumigatus, the most common cause of invasive pulmonary aspergillosis (IPA), produces a potent cytotoxjn called restrictocin. To investigate the role of restrictocin in (PA, we have constructed fungal strains in which the res gene has been inactivated by gene disruption. These disruptants lack the specific extracellular ribonucleolytic activity associated with restrictocin, as measured by an in vitro rabbit reticulocyte lysate assay. Western blot analysis of one drsruptant, using an anti-restrictocin monoclonal antibody, confirmed that the toxin is not produced. The growth characteristics of the disruptants could not be distinguished from those of their parental isolates on a variety of culture media. The pathogenicity of two disruptants was assessed in a murine model of IPA. There were no significant differences in mortality when these strains were compared with the parental isolates and an ectopic transformant. In addition, histological examination of infectediung tissue did not reveal any obvious differences in the number or size of fungal colonies or in the polymorphonuclearleucocyte response. Our results demonstrate that restrictocin is not an important virulence factor in this model of IPA.

    Notes: Summary The mineral apposition rate of trabecular bone was determined in several skeletal sites of young adult beagles. Tetracycline derivatives were administered intravenously or orally on 2 separate occasions preceding the day of sacrifice in order to label actively mineralizing bone surfaces. The rate of mineral apposition was calculated by dividing the distance between the 2 tetracycline markers by the time interval between their administration. The lumbar vertebra, proximal humerus, and pelvis, each of which contains red marrow, were found to have a significantly higher (P〈0.001) rate of mineral apposition in trabecular bone than the skeletal sites containing yellow marrow—the proximal ulna and distal humerus. The mean apposition rate in the former 3 sites was 1.3±0.3µm/day, while that in the latter 2 was 0.9±0.2µm/day (uncorrected for plane of sectioning). It is tempting to speculate that this finding may be a consequence of differences in vascularity between red and yellow marrow.

    Notes: Abstract Prophylactic orthopaedic management of spinal deformities in non-ambulatory children with the Duchenne-type muscular dystrophy has been enhanced by a computer-aided digitiser system. Routine standardised spine radiographs are analysed using a microprocessor-controlled digitiser interfaced to a desktop computer with a refreshing type c.r.t. display and full graphics capabilities. Analysis includes computation of conventional two-dimensional orthopaedic parameters as well as spatial (three-dimensional) quantities describing the actual geometry of the spine curvatures. Three views of the spine, antero-posterior (a-p), lateral and apical, are plotted on the c.r.t. Other graphical presentations include rotation of the a-p view about the vertical axis into the lateral view as well as perspective views of the three-dimensional geometry of the spine curves. Digitising and analysis of a particular patient's spine radiographs take place in the clinic while the patient is being examined. Comparison with previous examinations of spine deformities may be done by superimposing graphic representations of the spine curves. Optional routines include a comprehensive review of previously stored data for that patient, review of current data for selected patient groups and statistical analysis of variables from any of the available data files. Additional data files include neurology, pulmonary function and orthotics service. 332 patients are currently being followed with documentation generated using the system described in this article.

    Notes: Summary Hepatopancreas from Acellus intermedius, a small freshwater isopod, was examined by light and electron microscopy. Of particular interest are the two cell types present in the hepatopancreas. Their ultrastructural features indicate that the major role of the large alpha cells is to absorb material from the lumen as well as secrete materials into the lumen while the major role of the small beta cells is to store the absorbed materials as well as break down stored materials before transport.
