    Publication Date: 2021-05-19
Description: Two-hundred and sixty five green tiger shrimp juveniles (Penaeus semisulcatus) with the average weight of 7-12g were collected from Helleh farms in Bushehr province and transported to Iran Shrimp Research Center of Bushehr in October, 2009. The juveniles were acclimated for two weeks. The experiment was designed in three treatments (named 3, 4 and 5) and two controls (named 1 and 2) in triplicate with 15 shrimp in each repetition prepared of glass aquarium. All the treatments and repetitions were exposed to Vibrio harveyi (NCBI: GU974342.1). The concentrations of the treatments were 108,106 and 104 CFU ml-1 in individual containers dedicated for each mentioned treatment (3, 4 and 5, respectively). The controls prepared with no any bacteria and fully filled with chlorinated and UV treated sea water were named 1 and 2 respectively. The hemolymph were withdrawn from abdominal segments of samples for measuring THC and TPC evaluation at designed hours (2, 6, 12, 24, 48, 96, 144, 192 and 240). The shrimp samples were also fixed in Davidson fixative for histopathological studies. The results showed that the difference of THC value between controls and group 3 during 12 till 96 hours of experiment was significant (P〈 0.05). The differences between groups 4 and control of THC value during 24 to 144 hours of experiment were also significant (P〈 0.05). There was no significant difference (P〉 0.05) between group 5 and control groups of THC. The data showed that differences of TPC value between control groups during 24 to 96 hours were significant (P〈 0.05), whereas the differences between controls with groups 4 and 5 during 48 to 144 hours and 192 hours, were significant (P〈 0.05) respectively. TPC and THC were observed with an increase in the concentration of bacteria and passing the time as inverse bell shape procedure. In histopathology, gills showed melanization and color changed to brown and black. The hepatopancreas cells revealed necrosis and vacuolization of B, E, R and F cells. The bolitas ball and bacterial colonization was observed in the intestine. Our results showed that Vibrio harveyi with 108 and 106 cell/ml decreased immunity factors such as THC and TPC. The histopathological changes increased with increasing the concentration of bacterial level. This finding can be used for assessing the health of shrimp culture and prevention of vibriosis.

Description: White Spot Virus (WSV) is currently the most serious viral pathogen of shrimp worldwide it causes mortality up to 100% within 7-10 days in commercial shrimp farms. Infected Indian white shrimp Fenneropenaeus indicus samples were collected from Guatr shrimp site in Sistan and Baluchestan province in south of Iran and WSV infection was confirmed by Nested PCR. WSV was isolated from infected shrimp samples by centrifugation and filtration and multiplied in crayfish by intramuscular inoculation, the isolated virus was called WSV/IRN/1/2010. In order to determine the dilution resulting in 90-100% mortality in Penaeus semiculcatus, diluted virus stock in steps from 100 till 105 times in sterile PBS was injected intramuscularly to 14 shrimps in each group. Also the virus stock was diluted in steps from 1/2 till 1/32 times in sterile PBS and injected intramuscularly in Astacus leptodactylus crayfish. Therefore the LD50 of live virus stock in Astacus leptodactylus and Penaeus semiculcatus crayfish were calculated by the Karber method 10 3.29 /ml and 10 5.35 /ml, respectively.

Description: During the period from August 1997 to March 1998, two thousand samples of cultured Penaeus semisulcatus postlarvae and subadults were collected from five hatcheries and 20 growout farms distributed in three provinces along the costs of Persian Gulf and the Oman Sea. Based on growth signs, LM and TEM histopathology, Hepatopancreatic Parvo-like Virus (HPV) were identified from the samples. The HPV particles most often appeared in spherical, but occasionally in angular forms. The particles in hepatopancreatocyte sections, averaged 22-24nm in diameter.

Description: The virucidal effect of pH and salinity on Penaeus monodon baculovirus (MBV) was investigated by water borne inoculated methods exposing PL12 Penaeus semisulcatus for 10 hours and observing for 10 days at temperature 28- 30°C. For this purpose, one thousand virus free PL12 of P. semisulcatus were divided into five groups and each group placed in a 10 L plastic basin containing chlorine-treated sea water at 35ppt. The salinity of basins water after 12 hours changed to 5, 10, 20, 30 and 40ppt using synthetic sea salt. To study the effect of pH, solution with various pH of 3, 5, 7, 8, 9 and 12 were prepared using IN NaOH and IN HCL to Na2HPO4 solution. For transmission of MBV, an aliquot of approximately 10 frozen infected larvae were used for each experiment by water born inoculated method. Ten hours after inoculation, the shrimp were removed from jar and introduced into another jar with the same salinity or pH. During 10 days experiment, the rate of infection (ROI), severity of infection (SOI) and accumulative mortality were varied among the MBV-infected. Penaeus mondon baculovirus was able to infect Penaeus semisulcatus at salinity ranging from 5-40ppt whereas at pH 3 and 12, MBV was completely inactivated MBV was not able to infect larvae at pH 5, 7, 8 and 9.

Description: In the present Shrimp industry Has become a huge industry . The shrimp culture in Bushehr province began with Penaeus semisulcatus and Fenneropenaeus indicus and with introducing Litopenaeus vannamei in Iran Was known as a culture species. The shrimp production rate in IRAN is about 23000 metric ton. Outbreak of Various diseases and Biosecurity are most important things to produce Specific Patogen Free(SPF) shrimp. In this regard, production of SPF shrimp is The main requirements of shrimp industry. The survey on growth and srvival larve of different generation of Litopenaeus vannamei project was carried out in 2013 and 2014.Broodstock selecting ,propagation, culture and shrimp generation Was performed in 2012 until 2014.In 2014 we produced 4300 pieces of prebroodstock with 22.38 gram and 86 percent survival rate after 120 days of culture.
