    Notes: Conjugates of the single-chain ribosome-inactivating protein gelonin with ligands that bind to cell surface molecules vary greatly in their cytotoxicity. Conjugates that are not endocytosed after binding to cells exhibit low cytotoxicity similar to that of free gelonin, while conjugates that are endocytosed demonstrate enhanced cytotoxicity relative to free gelonin. However, the number of internalized gelonin molecules needed to intoxicate cells to the same degree has been found to be similar for all conjugates and for free gelonin. The intracellular concentration of gelonin has to be between 2,000-10,000 molecules/cell to achieve a surviving fraction of 0.37. Our studies revealed the presence of three distinct categories of cell surface molecules, those that are efficient in mediating endocytosis of im-munotoxins, those that are only moderately efficient, and those that seem not to cause internalization of bound immunotoxins.

    Notes: The fine structure of the mature spermatozoa of the feather-wing beetle (Coleoptera: Bambara new species no. 2) is described. Mature spermatozoa are extremely large in proportion to the size of the beetle. Under a light microscope a distinct head, midbody, and tail region can be seen with external coils in both the head and tail regions. Although the beetles were collected and preserved for skeletal anatomical study and not for histological preservation, the substructure comprising the spermatozoa was sufficiently well-preserved for their uniqueness to be appreciated. Observations with an electron microscope show that the axoneme is not contained in the tail region but is threaded through the head coils and then tightly wrapped around the midbody, terminating at the end of the midbody region. Sections through the midbody reveal three distinct layers: an inner electron dense core, a middle very elaborate crystalline Jattice-like matrix, and an outer axonemal wrapping. The structures observed in the head and tail regions appear to be modifications of those present in the midbody region. The possible functional significance of these unusual structures is discussed.

    Notes: The sensitivity of bacterial species to the lethal effects of radiations and alkylating agents is related to the base composition of their DNA; high guanine and cytosine (GC) content confers increased sensitivity to X rays and mustards and increased resistance to ultraviolet. Preliminary results suggest that differences in average GC content of different markers in Bacillus subtilis DNA may be detectable by radiosensitivity analysis. Purine starvation, thymine starvation, or incubation with certain purine and pyrimidine analogues may alter bacterial radiation responsiveness, as reflected in changes in the slope and/or shoulder of dose-survival curves; the responses differ characteristically with respect to such parameters as time of onset, reversibility, relative expression with X rays versus ultraviolet, oxygen-dependence, and glycerol-dependence. Available evidence pertaining to the molecular mechanisms of these radiosensitization phenomena is reviewed.

    Notes: Specific absorption rate (SAR) and effective depths of heating patterns induced by a shortwave, pancake diathermy applicator in fat-muscle phantom are measured. Midplane partitions of polyethylene and silk screen with and without contact chemicals are used. Thermographically obtained SAR data show nearly the same value for silk-screen partitions with and without contact chemicals and slightly lower values with polyethylene partitions, provided that the partition midplanes are tightly pressed against each other. Thermometry data indicate that for low-power exposures the major error in thermographic measurements obtained after termination of heating is due to thermal diffusion and not evaporative cooling in the opened midplane of the phantom.

    Notes: In attempting to evaluate the mechanisms responsible for susceptibility to the inductive increase in splenic complement receptor-positive (CR+) cells following exposure to 2450-MHz microwaves, it was found that sensitivity to microwave-induced CR+cell increases was under genetic control. In particular, evidence was accumulated suggesting that regulation was under the control of a gene or genes closely associated with but outside of the mouse major histocompatibility complex (H-2). All responsive strains of mice tested were of the H-2k haplotype, while mice of the H-2a, H-2b, H-2d and H-2i5 haplotypes were refractory to the microwave-induced increases in CR+ cells. By utilizing certain H-2k strains of mice that were genetically unable to respond to endotoxin, we were able to show that these strains of mice responded to microwaves, but not to endotoxin, by increasing CR+ cells. Microwave-induced increases in CR+cells were not mimicked by the intraperitoneal injection of hydrocortisone. Athymic mice responded to microwave exposure, indicating that this event was not regulated by the T-cell population. Mice less than eight weeks old were found not to be susceptible to exposure to 2450-MHz microwaves. These studies indicate that microwaves do induce changes in the population of cells with specific cell-surface receptors, that susceptibility to these changes is under genetic control, and that it is unlikely that endotoxin, corticosteroids, or regulatory T cells play a significant role in the mechanisms regulating these increases.
