    Notes: Abstract A 7.4-kilobase (kb) DNA plasmid was isolated from Glomerella musae isolate 927 and designated pGML1. Exonuclease treatments indicated that pGML1 was a linear plasmid with blocked 5′ termini. Cell-fractionation experiments combined with sequence-specific PCR amplification revealed that pGML1 resided in mitochondria. The pGML1 plasmid hybridized to cesium chloride-fractionated nuclear DNA but not to A + T-rich mitochondrial DNA. An internal 7.0-kb section of pGML1 was cloned and did not hybridize with either nuclear or mitochondrial DNA from G. musae. Sequence analysis revealed identical terminal inverted repeats (TIR) of 520 bp at the ends of the cloned 7.0-kb section of pGML1. The occurrence of pGML1 did not correspond with the pathogenicity of G. musae on banana fruit. Four additional isolates of G. musae possessed extrachromosomal DNA fragments similar in size and sequence to pGML1.

    Notes: Abstract The genomic clone RG64, which is tightly linked to the blast resistance gene Pi-2(t) in rice, provides means to perform marker-aided selection in a rice breeding program. The objective of this study was to investigate the possibility of generating a polymerase chain reaction (PCR)-based polymorphic marker that can distinguish the blast resistant gene, Pi-2(t), and susceptible genotypes within cultivated rice. RG64 was sequenced, and the sequence data was used to design pairs of specific primers for (PCR) amplification of genomic DNA from rice varieties differing in their blast disease responsiveness. The amplified products, known as sequenced-tagged-sites (STSs), were not polymorphic between the three varieties examined. However, cleavage of the amplified products with the restriction enzyme HaeIII generated a polymorphic fragment, known as specific amplicon polymorphism (SAP), between the resistant and the susceptible genotypes. To examine the power of the identified SAP marker in predicting the genotype of the Pi-2 (t) locus, we determined the genotypes of the F2 individuals at this locus by performing progeny testing for the disease response in the F3 generation. The results indicated an accuracy of more than 95% in identifying the resistant plants, which was similar to that using RG64 as the hybridization probe. The identification of the resistant homozygous plants increased to 100% when the markers flanking the genes were considered simultaneously. These results demonstrate the utility of SAP markers as simple and yet reliable landmarks for use in marker-assisted selection and breeding within cultivated rice.

    Notes: Proton and Carbon-13 NMR spectra of ethyl α-benzoyloxymethylacrylate (E)-methyl methacrylate (M) copolymers were analyzed in terms of sequence distribution and stereoregularity of monomer units. The copolymers were prepared by free radical polymerization in benzene at 50°C. The methoxy region of the M proton signal resonance was found to be sensitive to the copolymer composition for M-centred sequences. The carbon-13 NMR spectra of the EM copolymers, in particular the carbonyl signal resonances of carbomethoxy and carboethoxy groups, are discussed in terms of M- and E-centred configurational sequences. The experimental values were in excellent agreement with those calculated taken into account the terminal copolymerization model and Bernoullian distribution of stereoregularity with the statistical parameters determined from reactivity ratios rE = 0.32 and rM = 1.34 and the coisotacticity parameters σMM = 0.22, σEE = 0.70, and σME = σEM = σ = 0.30. © 1997 John Wiley & Sons, Inc.     Notes: Morphology and tensile properties of several styrene-butadiene copolymer reinforced asphalts have been investigated. In all cases, the raw materials were commercial samples; for the styrene-butadiene copolymer, a branching (nonlinear) block polymeric chain of high molecular weight was used. On the other hand, the asphalt was a nonoxydized commercial sample. The raw materials were characterized prior to making the composite. The morphology and mechanical properties of composites obtained at different chemical compositions were determined, and a comparison of these experimental results to different models, according to the composite morphology, is shown. © 1995 John Wiley & Sons, Inc.

    Notes: Rheological properties of styrene-butadiene copolymer-reinforced asphalt were investigated. A percolation-like model was used to fit dynamical experimental data, and a comparison with the Kerner-Takayanagi model is presented. In addition, a frequency-composition-temperature correspondence principle is proposed. This correspondence principle allows the prediction of the rheological behavior of an asphalt-based composite within a wide range of compositions, provided a narrow composition range at different frequencies and temperatures is previously known. © 1996 John Wiley & Sons, Inc.

    Notes: Fourier transform Raman (FTR) spectroscopy employing near-IR laser radiation at 1.06 μm as the scattering source was used to obtain Raman spectra of some neat energetic materials and several propellant formulations containing those energetic materials. It is shown that only crystalline components of propellant formulations are easily detected by FTR spectroscopy. Also, FTR spectroscopy is shown to be a useful tool in determining the principle ingredient(s) in many non-colored propellant formulations.
