    Notes: Nyctotherus cordiformis and Opalinids infesting Rana pipiens were studied by microincineration and in stained control sections. A marked difference in concentration and distribution of mineral salts was noted within saprozoic and holozoic organisms. The endoplasm in the saprozoic protozoa was free from inorganic residue except for the ‘vegetative’ granules; the protoplasm of the holozoic infusorians consisted of a fine network of mineral salts. Detailed examinations suggested that the qualitative and quantitative differences might be correlated with diffusion of digestible material through the cuticle. The inorganic residue of the walls of vacuoles was relatively rich in iron.

    Notes: Opalinids from the rectum of Rana pipiens were isolated in large numbers and fixed in a mixture of nine parts of absolute alcohol and one part of neutral formalin. These were embedded in paraffin and incinerated according to the technique of Policard. Examination of the preparations by dark-field illumination showed that the micro-incinerated protozoa retained their distinctive morphological and cytological characters, for the mineral ash had the same topographical distribution as the cell inclusions in the control stained preparations.The coarse vegetative granules of the cytoplasm were preserved as distinct masses of ash which corresponded exactly to these bodies in the control preparations. The structure of the myonemata and the cilia was perfectly preserved. The chromatin material of the nucleus, more or less abundant, left little or no ash, which is quite in contrast to the findings of Scott with regard to amphibian and mammalian nuclei.

    Notes: A total of 901 cumulus-oocyte complexes (COCs) were collected from bovine ovaries obtained at a local abattoir. COCs randomly assigned to Treatment I (n=451), were cultured in TCM-199 + 10% fetal bovine serum (FBS) and hormones, while oocytes in Treatment II (n=450) were cultured in TCM-199 + 20% estrous cow serum (ECS). Assessment of maturation revealed that 91.3% (42/46) of oocytes in Treatment I had reached metaphase II of meiosis, which was greater (P 〈0.05) than the 73.3% (33/45) in Treatment II. Following in vitro fertilization, 203 oocytes from Treatment I were co-cultured on bovine granulosa cells (Treatment IA) while the remaining 202 oocytes were co-cultured on bovine oviductal cells (Treatment IB). Similarly, 203 oocytes from Treatment II were co-cultured on granulosa cells (Treatment IIA) or oviductal cells (Treatment IIB, n = 202). Co-culture was maintained for 8 days. The proportion of cleaved zygotes was higher (P 〈0.05) in Treatment IB (86.6%) compared to Treatments IA (78.8%), IIA (58.1%), and IIB (64.8%). The proportion of cleaved zygotes that progressed beyond the 16-cell stage was also greater (P 〈0.001) in Treatment IB (71.4%) compared to Treatments IA (50.0%), IIA (35.4%) and IIB (55.8%). Treatment IB also produced the highest proportion of blastocysts (P〈0.0001) (41.1%) versus 24.6% (IA), 11.3% (IIA) and 18.3% (IIB). The proportion of day 6 morulae that progressed to form day 8 blastocysts was similar for both co-culture treatments (IA, 70.1%; IB 70.2%; IIA, 51.5%; IIB 50.8%) and varied only between in vitro maturation groups. Results indicate that the combination of in vitro maturation in TCM-99 + 10% FBS and hormones followed by co-culture on oviductal monolayers is a superior system for the production of early bovine embryos.

    Notes: Hydrocortisone modulates the binding capacity of HeLa cells for 125I-labeled epidermal growth factor (EGF). A twofold increase in 125I-labeled EGF binding is observed within 24 hours after the addition of pharmacological concentration of hydrocortisone (5 × 10-8-1 × 10-6 M). This enhancement of binding is reversible, and occurs when the cells are cultured in either serum-supplemented or completely defined, serum-free, hormone-supplemented medium. Scatchard analysis of the binding data indicates that the number of 125I-EGF binding sites is increased, and that no appreciable change in the affinity of the EGF receptor for labeled EGF occurs. In the serum-free condition hydrocortisone stimulates the growth of HeLa cells, but we have observed no connection between this growth stimulation and the enhancement of EGF binding. The growth response to hydrocortisone is independent of EGF, and the concentration dependency of the growth response to EGF is unaltered by the addition of hydrocortisone to the medium. Hydrocortisone elicits the growth response at a concentration as low as 5 × 10-9 M, while a concentration higher than 5 × 10-8 M is required to affect the binding capacity for 125I-EGF. These effects are specific for glucocorticoid steroids. Similar concentrations of progesterone, testosterone, or estradiol produce no measurable response. Although the elevation of EGF receptor levels in the serum-supplemented medium is similar to that observed in the serum-free cultures, hydrocortisone is growth-inhibitory under these conditions. This growth inhibition occurs at pharmacological concentrations of hydrocortisone with a concentration dependency that is similar to that of the EGF receptor modulation.

    Notes: A serum-free medium for serial culture of baby hamster kidney cell line 21 (BHK-21) as container-adherent cells was developed. The medium is a 1:1 (v/v) mixture of Dulbecco's modified Eagle's medium and Ham's F-12 medium supplemented with fibroblast growth factor, fibronectin, insulin, oleic acid (preincubated with fatty-acid-free bovine serum albumin as carrier), and transferrin. The fibronectin was required for cell adherence, the other factors for optimal cell multiplication. When cell input was greater than about 1,900 cells/cm2, this serum-free medium supported cell multiplication at a rate approximately equal to the rate in medium with 10% serum. At lower cell input, growth in the serum-free medium was poor unless it was supplemented with serum-free medium which had been conditioned by BHK-21 cells. The conditioned medium contained a factor(s) which enabled or stimulated cell multiplication.

    Description: Magellan probes Venus'surface by 12.6-cm-wavelength vertical and oblique radar scattering and measures microwave thermal emission. Emissivity and root-mean-square slope maps between 330 deg and 30 deg E and 90 deg N and 80 deg S are dissimilar, although some local features are exceptions. Inferred surface emissivities typically are 0.85, but vary from 0.35 at Maxwell to 0.95 northeast of Gula Mons and other locations. Lowest emissivities appear in topographically high areas; this relation suggests that a phase change or differences in chemical weathering occur at about 6055-kilometer radius. Initial results indicate that there are significant variations in the surface scattering function.

    Description: The Magellan radar mapping mission is in the process of producing a global, high-resolution image and altimetry data set of Venus. Despite initial communications problems, few data gaps have occurred. Analysis of Magellan data is in the initial stages. The radar system data are of high quality, and the planned performance is being achieved in terms of spatial resolution and geometric and radiometric accuracy. Image performance exceeds expectations, and the image quality and mosaickability are extremely good. Future plans for the mission include obtaining gravity data, filling gaps in the initial map, and conducting special studies with the radar.

    Description: The shape of seven large domes on the plains of Venus, with volumes between 100 and 1000 cu km, is compared with that of an axisymmetric gravity current spreading over a rigid horizontal surface. Both the altimetric profiles and the horizontal projection of the line of intersection of domes on the SAR images agree well with the theoretical similarity solution for a newtonian fluid, but not with the shape calculated for a rigid-plastic rheology, nor with that for a static model with a strong skin. As a viscous current spreads, it generates an isotropic strain rate tensor whose magnitude is independent of radius. Such a flow can account for the randomly oriented cracks that are uniformly distributed on the surface of the domes. The stress induced by the flow in the plains material below is obtained, and is probably large enough to produce the short radial cracks in the surface of the plains beyond the domes. The viscosity of the domes can be estimated from their thermal time constants if spreading is possible only when the fluid is hot, and lies between 10(exp 14) and 10(exp 17) Pa s. Laboratory experiments show that such viscosities correspond to temperatures of 610 - 690 C in dry rhyolitic magmas. These temperatures agree with laboratory measurements of the solidus temperature of wet rhyolite. These results show that the development of the domes can be understood using simple fluid dynamical ideas, and that the magmas involved can be produced by wet melting at depths below 10 km, followed by eruption and degassing.

    Description: Radiothermal emission results obtained by the Magellan spacecraft during its first 8-month cycle of data taking, corresponding to one full rotation of Venus, are reported. The new Magellan data confirm the existence of regions with extremely low values of radiothermal emission. Possible mechanisms to explain the anomalous phenomena are an interaction with a single interface between the atmosphere and a high-dielectric permittivity medium versus volume scattering from the interior of a low-loss medium. The discrimination between these mechanisms requires careful observation of the polarized state of the radiothermal emission and that of the radar scattering.

    Description: During the first 8 months of the Magellan mission, the radar altimeter has made some three million measurements of the surface of Venus covering the latitude range from 85 deg N to 80 deg S. Methods involving range correlation, Doppler filtering, multiburst summation, and range migration are used to focus the observations and to achieve high surface resolution. Results are presented as maps of the global distribution of topography, meter-scale roughness, and power reflection coefficient. The results are similar to those reported in previous experiments (surface heights exhibit a unimodal distribution with more than 80 percent of the surface lying within 1 km of the 6051.84-km mean radius) but the higher resolution of the Magellan altimeter has disclosed several surprisingly steep features, e.g., the southwest face of the Maxwell Montes, the southern face of the Danu Montes, and the chasmata to the east of Thetis Regio, where average kilometer-scale slopes of greater than 30 deg are not uncommon. This conclusion is corroborated by close inspection of synthetic aperture radar imagery.
