    Notes: It is shown that the presence of 31-35 commonly measured volatile organic compounds (VOCs) in ground water can be detected with small error rates by using screening methods which analyze for a subset of such VOCs. A study of selected data sets indicates that analytical determinations of only from two to eight VOCs will suffice to detect 95% of all VOC hits. It is also shown that a serially optimal algorithm for selecting the VOCs for screening is very nearly as accurate as a globally optimal algorithm and much easier to implement. These conclusions are supported by empirical evidence from two drinking-water data sets and one hazardous waste site data set. Additional research areas are also outlined.

    Notes: Interleukin-1β converting enzyme (ICE) processes the inactive proIL-1β to the proinflammatory mature IL-1β. ICE belongs to a family of cysteine proteases that have been implicated in apoptosis. To address the biological functions of ICE, we generated ICE-deficient mice through gene targeting technology. ICE-deficient mice developed normally, appeared healthy, and were fertile. Peritoneal macrophages from ICE-deficient mice underwent apoptosis normally upon ATP treatment. Thymocytes from young ICE-deficient mice also underwent apoptosis when triggered by dexamethasone, gamma irradiation, or aging. ICE-deficient mice had a major defect in the production of mature IL-1β and had impaired IL-1α production on LPS stimulation in vitro and in vivo.     Notes: Histopathology, morphometry, tritiated thymidin incorporation and immunohistochemistry were studied in 221 esophageal biospies from subjects with cytologica hyperplasia in Linxian, China. A spectrum of 7 morphologic entities were found (1) normal/near normal (NN); (2) basal cell hyperplasia 0 (BHO); (3) simple hyperplasia (SH); (4) mixed basal and spinous cell hyperplasia (MBS); (5) basal cell hyperplasia 1 (BH1); (6) dysplasia (D); and (7) non-profilerative lesion (NP). Forty percent of the biospies had combinations of histologic types. The thickness of the epithelium was increased in SH, MBS, and BH1, but not in BHO and NP Elongation of papiallae was frequent seen in SH, MBS, BH1, and D. Papillary bleeding was very prevalent in the esophageal specimens studied. A variety of cellular changes were found in peripapillary areas especially when bleeding occurred. [3H]-thymidine labeling index was dramatically increased in the entire epithelium in dysplasia, and also increased in cell layer 3 of MBS, BH1 and D. Blood group antigen LeY and lectin WGA showed consistent positivity in cellular membranes of the squamous cells, and these changes ocuurred before gross morphologic alterations. These findings provide a hypothesis for the sequences of pathogenetic events leading to esophgeal carcinoma, and define each step with corresponding biomakers for cancer prevention studies. © 1992 Wiley-Liss, Inc.

    Notes: Rates of CO2 production from glucose and glutamine, intracellular metabolite levels, and release of metabolic end products into the culture medium were determined for 13 cultured cell lines, including a glycolysis-defective mutant. All the non-mutant lines synthesized pyruvate, lactate, alanine, proline, aspartate, and citrate, so that the metabolism of glucose and glutamine resulted mainly in the production of these compounds and only to a lesser extent in complete oxidation to CO2. These data and the pattern of metabolites produced by the mutant line were consistent with a model characterized by incomplete glutamine oxidation leading to end product accumulation. Multiple linear regression analysis identified the metabolite levels most highly correlated with the intracellular citrate level and with the amount of citrate released into the medium. The analysis also showed that the rates of CO2 production from glucose and glutamine were themselves positively correlated, suggesting that the oxidation of the two substrates is coordinately controlled under normal culture conditions.
