    Beschreibung: The low prices and its relatively low carbon intensity of natural gas have encouraged the coal replacement with natural gas power generation. Such a replacement reduces greenhouse gases and other emissions. To address the significant energy penalty of carbon dioxide (CO2) sequestration in gas turbine systems, a novel high efficiency concept is proposed and analyzed, which integrates a flame-assisted fuel cell (FFC) with a supercritical CO2 (sCO2) Brayton cycle air separation. The air separation enables the exhaust from the system to be CO2 sequestration-ready. The FFC provides the heat required for the sCO2 cycle. Heat rejected from the sCO2 cycle provides the heat required for adsorption-desorption pumping to isolate oxygen via air separation. The maximum electrical efficiency of the FFC sCO2 turbine hybrid (FFCTH) without being CO2 sequestration-ready is 60%, with the maximum penalty being 0.68% at a fuel-rich equivalence ratio (Φ) of 2.8, where Φ is proportional to fuel-air ratio. This electrical efficiency is higher than the standard sCO2 cycle by 6.85%. The maximum power-to-heat ratio of the sequestration-ready FFCTH is 233 at a Φ = 2.8. Even after including the air separation penalty, the electrical efficiency is higher than in previous studies.

    Beschreibung: Salmonella is a burden to the poultry, health, and food safety industries, resulting in illnesses, food contamination, and recalls. Salmonella enterica subspecies enterica Enteritidis (S. Enteritidis) is one of the most prevalent serotypes isolated from poultry. Salmonella enterica subspecies enterica Heidelberg (S. Heidelberg), which is becoming as prevalent as S. Enteritidis, is one of the five most isolated serotypes. Although S. Enteritidis and S. Heidelberg are almost genetically identical, they both are capable of inducing different immune and metabolic responses in host cells to successfully establish an infection. Therefore, using the kinome peptide array, we demonstrated that S. Enteritidis and S. Heidelberg infections induced differential phosphorylation of peptides on Rho proteins, caspases, toll-like receptors, and other proteins involved in metabolic- and immune-related signaling of HD11 chicken macrophages. Metabolic flux assays measuring extracellular acidification rate (ECAR) and oxygen consumption rate (OCR) demonstrated that S. Enteritidis at 30 min postinfection (p.i.) increased glucose metabolism, while S. Heidelberg at 30 min p.i. decreased glucose metabolism. S. Enteritidis is more invasive than S. Heidelberg. These results show different immunometabolic responses of HD11 macrophages to S. Enteritidis and S. Heidelberg infections.

    Beschreibung: Yersinia entercolitica is a bacterial species within the genus Yersinia, mostly known as a human enteric pathogen, but also recognized as a zoonotic agent widespread in domestic pigs. Findings of this bacterium in wild animals are very limited. The current report presents results of the identification of cultures of Y. entercolitica from dead bats after a massive bat die-off in a cave in western Georgia. The growth of bacterial colonies morphologically suspected as Yersinia was observed from three intestine tissues of 11 bats belonging to the Miniopterus schreibersii species. These three isolates were identified as Y. enterocolitica based on the API29 assay. No growth of Brucella or Francisella bacteria was observed from tissues of dead bats. Full genomes (a size between 4.6–4.7 Mbp) of the Yersinia strains isolated from bats were analyzed. The phylogenetic sequence analyses of the genomes demonstrated that all strains were nearly identical and formed a distinct cluster with the closest similarity to the environmental isolate O:36/1A. The bat isolates represent low-pathogenicity Biotype 1A strains lacking the genes for the Ail, Yst-a, Ysa, and virulence plasmid pYV, while containing the genes for Inv, YstB, and MyfA. Further characterization of the novel strains cultured from bats can provide a clue for the determination of the pathogenic properties of those strains.

    Beschreibung: With the reemergence of poultry diseases such as necrotic enteritis following the restriction of in-feed antibiotics, the search for antibiotic alternatives has become critically important. Postbiotics are non-viable bacterial products or metabolic byproducts from probiotic microorganisms that have positive effects on the host or microbiota. These are a promising alternative to antibiotics. Here, we describe the mechanism of action of a postbiotic in the context of a Clostridium perfringens (C. perfringens) challenge model. By using performance measurements and a peptide array kinome analysis, we describe the kinotypes and signal transduction changes elicited by the postbiotic with and without C. perfringens challenge. The postbiotic improves lesion scores, C. perfringens counts and mortality compared to challenge groups without the postbiotic, and it improves weight gain in the most severely challenged birds. The postbiotic predominantly affects the innate immune response and appears immunomodulatory. In the context of infection, it reduces the proinflammatory responses and generates a homeostatic-like response. This postbiotic is a viable alternative to antibiotics to improve poultry health in the context of C. perfringens pathogen challenge.

    Beschreibung: Wastewater treatment plant (WWTP) utilization of combined heat and power (CHP) systems allows for the efficient use of on-site biogas production, as well as increased annual savings in utility costs. In this paper, a review of biogas energy recovery options, CHP prime mover technologies, and the costs associated with biogas cleaning give a broad summary of the current state of CHP technology in WWTPs. Even though there are six different prime mover technologies, the main ones currently being implemented in WWTPs are micro turbines, fuel cells and reciprocating engines. Different prime movers offer varying efficiencies, installation costs, and biogas impurity (H2S, siloxanes, HCl) tolerances. To evaluate the long-term savings capabilities, a techno-economic assessment of a CHP installation at a case study WWTP shows the payback, annual savings, and initial costs associated with the installation of a CHP system. In this case, a study a payback of 5.7 years and a net present value of USD 709,000 can be achieved when the WWTP generates over 2,000,000 m3 of biogas per year and utilizes over 36,000 GJ of natural gas per year.

    Beschreibung: Metal pollution is pervasive across terrestrial and aquatic ecosystems owing to anthropogenic activities. Sediments can accrue high concentrations of metals and act as secondary sources, and thus may be valuable indicators of metal contamination across spatiotemporal scales. In aquatic systems, the extent of metal pollution may be further mediated by transference among sediments and living organisms, with plant metal contaminants potentially predictive of underlying sediment concentrations. The present study thus quantifies the extent of metal pollutants (Na, K, Ca, Mg, Cu, Zn, Mn, B, Fe) across multiple study sites and seasons (cool-dry, hot-wet, hot-dry) in a subtropical river system. Furthermore, uptake by a key macrophyte species, Phragmites australis, was examined and correlated with sediment pollution levels among different plant parts. Overall, sediment pollution load indices differed seasonally, being significantly highest during the cool-dry season irrespective of sampling location, suggesting that periods with reduced water flows can exacerbate metal pollution levels in riverine sediments. Also, metal concentrations were highest in upstream wetland sites, indicating a capacity for metal sink effects in these areas. Overall, macrophytes contained high concentrations of select metals, however composition and concentrations differed across plant parts, with roots containing particularly high concentrations of Fe and B. Correlations between sediment and macrophyte concentrations were mostly non-significant, whilst stem Mn and Fe concentrations correlated significantly negatively and positively to sediment concentrations, respectively. The present study identifies key spatiotemporal differences in multiple metal contaminants in an understudied subtropical aquatic system that align with hydrological regime differences. Whilst macrophytes were not found to be major accumulators, or predictors, of metal contaminants in this study, they may collectively play a central role in concentration regulation in aquatic systems.

    Beschreibung: Dictyostelium discoideum is gaining increasing attention as a model organism for the study of calcium binding and calmodulin function in basic biological events as well as human diseases. After a short overview of calcium-binding proteins, the structure of Dictyostelium calmodulin and the conformational changes effected by calcium ion binding to its four EF hands are compared to its human counterpart, emphasizing the highly conserved nature of this central regulatory protein. The calcium-dependent and -independent motifs involved in calmodulin binding to target proteins are discussed with examples of the diversity of calmodulin binding proteins that have been studied in this amoebozoan. The methods used to identify and characterize calmodulin binding proteins is covered followed by the ways Dictyostelium is currently being used as a system to study several neurodegenerative diseases and how it could serve as a model for studying calmodulinopathies such as those associated with specific types of heart arrythmia. Because of its rapid developmental cycles, its genetic tractability, and a richly endowed stock center, Dictyostelium is in a position to become a leader in the field of calmodulin research.

    Beschreibung: Transition metals, including copper, iron, and manganese, are known to catalyze the generation of reactive oxygen species (ROS) in beer leading to reduced product stability. Metals in beer are generally derived from raw ingredients. The present study aims to evaluate the impact of brewing and dry-hopping using hops treated with copper-based fungicides (CBFs) on the final transition metal content of model buffer solutions and pilot-scale systems of wort and beer. Copper levels in model wort and beer solutions were elevated (105.6% and 230.4% increase, respectively) when CBF-treated hops were used. In laboratory-prepared wort, elevated copper concentrations were not observed when CBF-treated hops were used for boiling. Dry hopping of beer using CBF-treated hops led to significant increases in total copper content (ca. 75 µg/kg vs. ca. 40–50 µg/kg in the control-hopped beer) when yeast was absent from the treated beer, but not when yeast was present. It was observed that manganese levels were significantly elevated in all hopped beers (ca. 495–550 µg/kg vs. 90–125 µg/kg in the unhopped control), regardless of hop treatment. A hop varietal thiol, 4-Mercapto-4-methylpentan-2-one, was spiked into treated beers, and the rate of oxidative loss was monitored during aging. Rates of thiol loss in treated beer samples did not differ across CBF treatments but were significantly lower in unhopped controls in the absence of yeast (p 〈 0.0001) and correlated significantly with total manganese content of the beers (R2 = 0.4228, p = 0.0006). The rate of staling in hopped beers as measured by the rate of 1-hydroxyethyl radical generation did not differ among hop treatments, suggesting that excess copper content contributed from the hops does not negatively impact the oxidative stability of the beers. These findings suggest that brewers can use CBF-treated hops without any negative implications for the shelf stability of their beers and do not contraindicate the use of CBF in hops production when necessary.

    Beschreibung: Elytroteinus geophilus (Lucas) is a polyphagous weevil that is widespread in the South Pacific islands and is known to cause damage to various crops with large nutrient storage structures such as kava, ginger, yellow passion fruit, yams and sweet potatoes. More significantly, E. geophilus, the Fijian Ginger Weevil, has been linked, along with two wound invading fungal pathogens, to a passion fruit collar rot in Samoa. This species is considered a high-risk insect pest and it is included in the USDA’s prioritized offshore pest list. We report on new plant hosts and behavior of this weevil. The first new host record resulted from interception of this weevil in bulbs of Tropical Spider Lily (Crinum sp.) in Alabama. This interception initiated an examination of museum specimens and the literature that resulted in a second previously unreported host record (vanilla (Orchidaceae)) and a new behavioral trait for this weevil: the use of plant fibers to spin a cocoon for pupation. A synthesis of known host plants records is reported here and suggests a preference by this weevil of starch-storing plant organs. A distribution map and a differential diagnosis of the species is also provided.

    Beschreibung: Drug delivery to the anterior and posterior segment of eye remains a challenge. Nanoparticle-mediated drug delivery has indicated some promise. The presented review aims to summarize recent advancements in chitosan-based nanotherapies for ocular drug delivery and the challenges encountered during the process. Significant research using chitosan, a cationic linear polymer, is being conducted for ocular drug delivery. A vast number of publications exploit the mucoadhesive properties of the polymer, which arise due to interactions between the amino acids of chitosan and the sialic acid residues in mucous. The high degree of crosslinking in chitosan nanoparticles facilitates a dramatic increase in ocular drug retention of the desired drug, which subsequently helps in ocular penetration and improving the bioavailability of the drugs. A noted decrease in the initial burst of the drug is the basis for developing sustained drug release formulation using biodegradable and biocompatible chitosan polymer. In vitro as well as in vivo studies have indicated enhancement in the uptake, accumulation, and removal of chitosan nanoparticles from the site of delivery. In summary, chitosan- or modified-chitosan-based nanoparticles are being widely tested as drug carriers for treatment of bacterial and viral infections, glaucoma, age-related macular degeneration, and diabetic retinopathy.
