    Notes: In this work we explore the possibilities of the gel-acupuncture technique, proposed previously for the growth of protein single crystals [García-Ruiz, Moreno, Viedma & Coll (1993). Mater. Res. Bull. 28, 541–546]. The main advantage of the technique is that the crystals are obtained inside an X-ray capillary and, unlike classical microdiffusion techniques, it involves a very simple and accurate technical arrangement that permits the continuous monitoring of the crystals in their growth environment. In particular, we describe the growth of single crystals of lysozyme, concanavalin A and ribonuclease A. Different starting conditions have been used to grow single crystals of these proteins into different types of capillaries at several protein and precipitating-agent concentrations. It is demonstrated that the technique works for a wide range of precipitating agents commonly used in protein crystal growth, such as large polymers (PEG 4000 and PEG 6000), organic solvents (from methanol to butanol) and salts [NaCl, (NH4)2SO4]. The range of inner diameter of the capillaries for which the technique works correctly has been also studied. The growth process and possible crystal movement was followed by video microscopy. Lysozyme crystals up to 3.1 mm were obtained but the average maximum linear crystal sizes were 2.0 mm for lysozyme, 0.4 mm for concanavalin A and 1.2 mm for ribonuclease, respectively. The waiting times to reach such a size, measured from the set-up of the experiments, were 72 h, 10 d and 5 d, respectively. Gels of tetramethoxysilane, tetraethoxysilane, sodium silicate, agar, high-strength agar and gel-gro have been tested in relation to their mechanical properties and their chemical interaction with the reactants. Finally, we discuss briefly the advantages of the gel-acupuncture technique and plausible applications other than crystal growth.

    Notes: C-phycocyanin and allophycocyanin from the green alga Spirulina platensis were isolated and crystallized by gel-acupuncture techniques. A novel two-step chromatographic procedure was used for purification. Blue hexagonal crystals were obtained by diffusing magnesium chloride into the protein solution for a week, followed by diffusion of PEG 6000 in order to complete the reduction of the solubility of the protein in the capillary tube used as a growth cell. In the case of allophycocyanin, crystals with a size of 0.4 × 0.3 × 0.3 mm were characterized by X-ray diffraction. They belong to space group P6322 with unit-cell parameters a = b = 102.04, c = 131.22 Å. The crystals of C-phycocyanin belong to either space group P6 or P63 with unit-cell constants a = b = 182.38, c = 60.87 Å, α = β = 90, γ = 120°. The crystals diffract beyond 2.4 and 2.5 Å resolution, respectively, using a rotating anode as an X-ray source.

    Notes: Abstract  Sequences of Aotus nancymaae immunoglobulin kappa light-chain rearrangements were analyzed after reverse transcription polymerase chain reaction. Among 22 in-frame rearrangements analyzed, 12 IGKV genes belonging to the families 1, 2, or 3 were identified. Aotus counterparts for all five human IGKJ genes were found. The identity of the deduced human and Aotus amino acid sequences was between 83% and 92% for junctional regions and 74% for the constant region. Sequence comparisons between rearrangements indicated that somatic mutations, the addition of nongermline-encoded nucleotides, and exonuclease trimming contribute to the generation of diversity of Aotus immunoglobulin kappa chains. The high identity of Aotus and human IGK genes is comparable to that of T-cell receptor genes and further supports the proposal to use the Aotus Plasmodium falciparum infection model for the evaluation of malaria vaccine candidates.

    Notes: Abstract  The structure and seasonal changes of earthworm communities were evaluated in a natural savanna and in a improved grass-legume pasture in a Colombian oxisol over a period of 18 months. One plot of 90×90 m was isolated in each of the systems and each month five samples of 1 m2×0.5 m and ten of 20×20×20 cm were randomly selected from a stratified block design. Species richness was similar in the two evaluated plots (seven species), whereas diversity measured by the index, H (Shannon and Weaver 1949) was clearly different, i.e. H=2.89 in natural savanna and H=1.29 in pasture. This is explained by differences in earthworm community structure. The average annual density in the savanna was 49.8, ranging from 10.8 to 135.8 individuals (ind) m–2, and biomass was 3.3 g m–2 (hand-sorting method), ranging from 0.9 to 11.5 g m–2. In the man-made pasture, density was 80.1 ind m–2 on average, ranging from 24 to 215.8 ind m–2 and biomass was more than tenfold higher, ranging from 29.2 to 110.4 g m–2. This was especially due to the presence of a large glossoscolecid anecic species, Martiodrilus carimaguensis Jiménez and Moreno, which has been greatly favoured by conversion of savanna to pasture. Endogeic species were dominant in the natural savanna whereas the anecic species accounted for 88% of total earthworm biomass in the pasture. Total earthworm density and biomass were significantly different in the two systems studied (t-test). The results indicate a clearly positive response of earthworm communities to improved pastures, a type of land use that is being increasingly adopted in moist neotropical savannas.

    Notes: Abstract To study the titanium effect on P nutrition, a greenhouse experiment withCapsicum annuum L., cv. Bunejo plants growing under differential P fertilization was conducted. All the plants were grown under identical conditions and they only differred in the P fertilization and in Ti supply. Plant biomass production of the Ti-untreated plants was affected by the diminution of the P-feed, but the plants growing under the lowest P supply did not showed any deficiency symptom during the crop cycle. All the Ti-treated plots showed a significative increase of the plant biomass against their corresponding untreated references. The biomass enhancement was mainly caused by the increase of the fruit yield with an absolute enhancement of 62% in the plants growing under the lowest P feed, and of 45% in the plants with a complete P support.

    Notes: Abstract Objectives: The aim of this study was to characterize the use of seralbumin, evaluating how appropriate its prescription is and what possible economic repercussions may result from inappropriate use. Methods: We performed a prospective study that included all patients receiving albumin in two University Hospitals from October 1995 to March 1996. The reasons for albumin use were considered appropriate if they coincided with the recommendations of a panel of experts. Results: During the study period, 197 patients received albumin and a total of 3208 50-ml vials (20%) were used. The internal medicine and gastroenterology services prescribed this drug the most often. The most frequent prescription motives were paracentesis in cirrhotic patients (25.9%), hypoalbuminemia (24.9%) and chronic handling of cirrhotic patients (18.6%). Only 16 prescriptions (8.1%) (corresponding to 315 vials, 9.8%) were considered appropriate. One cause of inappropriate prescribing was that colloid solutions had not previously been used in 56 (30.9%) of the 186 inappropriate prescriptions. During the study period, 74 306 ECUs were spent on inappropriate indications. Conclusions: The use of albumin in our centers is incorrect and has important economic repercussions. Some educational and informative measures must be established to change this situation.

    Notes: Abstract The stress corrosion cracking is a typical fracture process in metals and alloys. Among aluminium alloys, the 7075 alloy, presents a high performance in the mechanical properties but it is susceptible to stress corrosion cracking. This paper presents a semiempiric model of crack growth by stress corrosion cracking for the above alloy. This model only uses macroscopic parameters from fracture mechanic theory and experimental tests which are easy to obtain. The model quantifies the fissure rate related to environmental condition, microstructure and loading level, permitting the evaluation of the crack growth process at different environmental conditions and heat treatments. The model results are compared with the experimental data obtained. The theoretical model reproduces adequately the stress corrosion cracking process for the 7075 alloy.
