Notes: Bench scale packed columns were used to study the kinetic behavior of biological denitrification. After tracer studies were run to determine flow characteristics, steady state nitrate profiles were determined. A first-order rate expression with an Arrhenius temperature dependence is an adequate model of the reaction for nitrate-nitrogen concentrations of 〈 100 mg/l and temperature of 5° to 20°C. The rate constants are proportional to the packing surface area.

Notes: Infeasible path optimization algorithms have been effective with equation-solving or simultaneous modular process simulators. Using these simulators, successive quadratic programming is applied to large nonlinear programs. However, the algorithms are not directly applicable to commercial simulators of the sequential modular type. Here an infeasible path algorithm is presented which can easily be integrated with most commercial process simulators. The optimization problem remains small (10 or 20 variables) and the user merely replaces the convergence block with an “optimization” block.A simple process example demonstrates the operation, effectiveness, and potential of this algorithm.

Notes: The plastics industry is now coming to realize that copolymers are unique thermoplastics which have broad areas of application. It is believed that ethylene-methyl acrylate copolymers of 2 melt index may be extrusion coated to paper by a cap-coat process or by use of a chill roll with an adhesive coating. Copolymers of ethylene and vinyl monomers can be made into film by the same methods used for polyethylene. Methyl acrylate and methyl methacrylate copolymers may be processed on a wide range of molding equipment. Table VII in the article gives a list of applications for each polymer, along with expected physical properties.

Notes: A β-galactoside-binding endogenous lectin extracted from bovine heart binds to the surface of baby hamster kidney (BHK) cells. The binding to and agglutination of cells is reduced in certain ricin-resistant mutants (Ric cells) in parallel with the decreased number of binding sites for the selective agent, ricin, a galactose-specific plant lectin. However, clear differences in the binding specificities of bovine lectin and ricin are shown by the effect of neuraminidase. BHK cells and Ric mutant cells treated with neuraminidase bind similar amounts of the bovine lectin compared with untreated cells, and ricin binding is greatly increased. The mammalian lectin immobilised on inert glass mediates the attachment and spreading of normal BHK cells and agglutinates these cells in solution. Ricin-resistant mutant cells respond poorly. These results are consistent with a role of endogenous lectins in cellular adhesiveness and show that cell adhesion may be regulated by the density of specific surface receptors for lectins.

Notes: In the present study the conditions leading to tight binding of human serum albumin to the Amberlite XAD-7 resin without the use of chemical coupling agents have been defined. Optimal binding (10.97 mm/kg dry XAD-7) was achieved at pH 5.0 and adsorption conformed to a Langmuir isotherm. Theoretical analysis of the data suggest adsorption of a monolayer of albumin which is suported by the absence of visual surface coating on scanning electron micrographs. Binding of human serum albumin was reduced when two chemical coupling agents, glutaraldehyde and carbodiimide were included in the human serum albumin solution, the elution of adsorbed HSA from the resins under severe flow conditions was reduced, and the amount eluted was minimal in all instances.

Notes: In the light of accumulating data that implicate cell surface heparan sulfate proteoglycans (HSPGs) with a role in cell interactions with extracellular matrix molecules such as fibronectin, we have compared the properties of these molecules in wild-type BHK cells and an adhesion-defective ricin-resistant mutant (RicR14). Our results showed that the mutant, unlike BHK cells, cannot form focal adhesions when adherent to planar substrates in the presence of serum. Furthermore, while both cell lines possess similar amounts of cell surface HSPG with hydrophobic properties, that of RicR14 cells had decreased sulfation, reduced affinity for fibronectin and decreased half-life on the cell surface when compared to the normal counterpart. Our conclusions based on this data are that these altered properties may, in part, account for the adhesion defect in the ricin-resistant mutant. Whether this results from the known alteration in assembly of N-linked glycans affecting the carbohydrate chains on the proteoglycan or some other combination of factors is discussed.
