    Description: Ecological and socioeconomic impacts from biological invasions are rapidly escalating worldwide. While effective management underpins impact mitigation, such actions are often delayed, insufficient or entirely absent. Presently, management delays emanate from a lack of monetary rationale to invest at early invasion stages, which precludes effective prevention and eradication. Here, we provide such rationale by developing a conceptual model to quantify the cost of inaction, i.e., the additional expenditure due to delayed management, under varying time delays and management efficiencies. Further, we apply the model to management and damage cost data from a relatively data-rich genus (Aedes mosquitoes). Our model demonstrates that rapid management interventions following invasion drastically minimise costs. We also identify key points in time that differentiate among scenarios of timely, delayed and severely delayed management intervention. Any management action during the severely delayed phase results in substantial losses (〉50% of the potential maximum loss). For Aedes spp., we estimate that the existing management delay of 55 years led to an additional total cost of approximately $ 4.57 billion (14% of the maximum cost), compared to a scenario with management action only seven years prior (〈 1% of the maximum cost). Moreover, we estimate that in the absence of management action, long-term losses would have accumulated to US$ 32.31 billion, or more than seven times the observed inaction cost. These results highlight the need for more timely management of invasive alien species—either pre-invasion, or as soon as possible after detection—by demonstrating how early investments rapidly reduce long-term economic impacts.

    Notes: Abstract A study has been made of the electrochemical reduction of pentachlorophenol (PCP) on a lead electrode in both water and propylene carbonate. A combination of voltammetric measurements and analysis of reaction products by high performance liquid chromatography (HPLC) have established that the reduction of PCP in propylene carbonate occurs by two different pathways, depending upon the electrode potential. At -1.9V vs SCE, reduction is mediated by adsorbed hydrogen, whereas at -2.3V vs SCE direct reduction of PCP also occurs. A similar approach has established that the reduction of PCP in water follows only one reaction pathway, namely, direct electron transfer from lead to PCP.

    Notes: Summary We have used a PCR-based technique, involving the random amplification of polymorphic DNA (RAPD), to assess genome variability between 21 isolates from F. solani f. sp. cucurbitae races 1 and 2. Based on RAPD marker patterns the isolates fell into two distinct groups corresponding to mating populations MPI and MPV. Four isolates that could not be assigned to one or other mating population by traditional means were distinguished by RAPD patterns. Seven polymorphic RAPD products were used to probe Southern blots of MPI and MPV genomic DNA. Six of the seven probes hybridized to single-copy sequences and five of the seven probes showed specificity for one or other mating population. We suggest that not only is the technique a rapid and reliable tool for isolate-typing of fungi but it also provides a rapid method for obtaining species- or racespecific hybridization probes.

    Notes: Abstract The phase transition between the $$P\bar 1$$ and $$I\bar 1$$ phases of anorthite has been studied at elevated pressure by single-crystal X-ray diffraction in a diamond-anvil cell. The phase transition is shown to be first-order in character for both end-member anorthite (CaAl2Si2O8) and for an anorthite with a small amount of albite component (NaAlSi3O8) in solid solution. Reversals of the transition across the phase boundary at three other compositions show that the transition pressure (P Tr) increases with increasing albite content. This behaviour is compared with that observed at elevated temperatures, and is analysed in terms of Landau theory.

    Notes: Abstract High-pressure single crystal X-ray diffraction experiments of phase anhydrous B and superhydrous B have been carried out to 7.3 and 7.7 GPa, respectively, at room temperature. Fitting a third-order Birch-Murnaghan equation of state to the P-V data yields values of V 0 = 838.86 ± 0.04 Å3, KT,0 = 151.5 ± 0.9 GPa and K′ = 5.5 ± 0.3 for Anhy-B and V 0 = 624.71 ± 0.03 Å3, KT,0 = 142.6 ± 0.8 GPa and K′ = 5.8 ± 0.2 for Shy-B. A similar analysis of the axial compressibilities in Anhy-B reveals that the c-axis is most compressible (Kc = 137 ± 3 GPa), the b-axis is least compressible (Kb = 175 ± 4 GPa), and the a-axis is intermediate (Ka = 148 ± 1 GPa). In Shy-B, the a-axis is most compressible (Ka = 135 ± 1 GPa), followed by the b- and c-axes which have similar compressibilities (Kb = 146 ± 3 GPa; Kc = 148 ± 3 GPa). The fact that the b-axis of Shy-B is approximately 16% more compressible than Anhy-B is primarily due to differences in the O-T layer in which the H atoms are located and the linkages with the adjacent O layers. The rigid edge-sharing chains of MgO6 and SiO6 octahedra in the O layer control compressibility along the a- and c-axes in both structures. The net result is a reduction in the overall anisotropic compression from ∼22% in Anhy-B to ∼9% in Shy-B.

    Notes: Abstract An homologous transformation system has been developed for the plant pathogenic fungus Glomerella cingulata (Colletotrichum gloeosporioides). A transformation vector containing the G. cingulata gpdA [romoter fused to the hygromycin phosphotransferase gene was constructed. Southern analyses indicated that this vector integrated at single sites in most transformants. A novel method of PCR amplification across the recombination junction point indicated that the integration event occurred by homologous recombination in more than 95% of the transformants. Deletion studies demonstrated that 505 bp (the minimum length of homologous promoter DNA analysed which was still capable of promoter function) was sufficient to target integration events. Homologous integration of the vector resulted in duplication of the gdpA promoter region. When transformants were grown without selective pressure, a high incidence of vector excision by recombination between the duplicated regions was evident. The significance of these recombination characteristics is discussed with reference to the feasibility of performing gene disruption experiments.

    Notes: Summary A cosmid vector, suitable for library construction and DNA transformation in filamentous fungi, has been constructed and a reliable and highly efficient PEG-mediated DNA transformation system for F. solani f. sp. cucurbitae, based on resistance to hygromycin B, has been developed for use with this vector. This transformation system yielded 104 transformants per μg of DNA when using 107 protoplasts. Factors important in achieving high efficiency included: the maintenance of an osmoticum in all transformation steps, PEG 4000 concentration, and the ratio of transforming vector DNA to protoplasts. Approximately 60% of transformants stably integrated vector DNA. Molecular analysis revealed multiple copies of the plasmid integrated into the genome at one or more sites. The frequency of transformation achieved will facilitate the isolation of genes from this fungus by complementation.

    Notes: Abstract In tomato, resistance to the wilt fungus Verticillium albo-atrum is determined primarily by the Ve locus. When two tomato near-isolines which differ at this locus and in their susceptibility to the pathogen were compared, more rapid suberin coating in the xylem of resistant plants correlated closely with a more rapid increase in the activity of phenylalanine ammonia-lyase (PAL; EC 4.3.1.5), an enzyme which is essential to the suberization process. In contrast, levels of mRNA did not increase proportionally to the measured enzyme activities; rather, there was a substantial suppression of mRNA levels in the susceptible tomato line, consistent with a much lower elevation of PAL activity and significantly less vascular coating. The suppression was absent or substantially reduced in the resistant line. The results indicate that the pathogen can suppress defense genes in susceptible plants but suggest that their expression is altered in resistant hosts and that post-transcriptional regulation plays a significant role.
