    Notes: Summary The relationship of diet and phylogeny to the intrinsic rate of population increase (r max) was examined in a sample of 39 mammalian species that live in Neotropical forests. Diets of species did not predict their r max, contrary to published predictions based on associations between basal metabolic rate and diet and between basal metabolic rate and r max. Phylogeny did however, apparently because life history characteristics and susceptibility to predation vary predictably with phylogeny and with one another.

    Notes: Abstract Samples taken at scattered points of a finite-support two-dimensional signal can be interpolated to recover an approximation of the original signal. Given a bound on the number of samples, where should they be placed to enable the most accurate reconstruction? Or, given an error bound for the reconstruction, what is the minimum number of samples required, and where should they be placed? In this paper we introduce search schemes that provide good candidate solutions to these problems, for digital signals. Natural Neighbour Interpolation is used in iterative sample removal and movement processes to obtain sparse sample patterns. For pictures and Digital Elevation Models, fewer samples are required if the interpolant is onlyC 0 continuous at the data sites, than if it isC 1. Retained samples lie on the ridges and valleys of the laplacian.

    Notes: Abstract A common view of the energy problem holds that what is at stake in most energy policy debates are questions of fact that are in principle susceptible to resolution by objective, scientific research. It is argued in this paper that this view is misguided, and that underlying many of the factual issues apparently in dispute are differences at the framework level, that is differences in basic presuppositions and the patterns of thinking employed. By means of an examination of the debate over soft and hard energy paths, it will be argued that framework differences are fundamental to that debate and that such differences are not susceptible to factual resolution. As a result, the debate has taken on the character of a ritualized performance, as each side strives to convince, not their opponents, but policymakers of the rightness of their opinion. Some of the implications of these findings for energy policymaking are explored.

    Notes: Abstract This article examines two examples of what has been called mandated science - risk analysis and long-term energy demand forecasting - with a view to disentangling the implicit assumptions underlying their use in public policy debates and in the policy process. In both fields, there has emerged a growing and very similar challenge to traditional views of the role and status of mandated science in informing decision-making. This challenge has led to the reformulation of some of the attributes of the standard view, without however causing changes in the underlying view of the nature of the problem or the role of science in principle. A deeper critique suggests that mandated science is inherently value-laden, producing conclusions that are strongly affected by theoretical presuppositions that in turn reflect underlying values (e.g., about rationality, knowledge or the role of technology). Thus a sharp separation of science and policy is untenable and amounts to a hidden policy bias in favor of the business-as-usual policies supported by conventional science. There is a need for a new concept of science, one that is more integrated into the policy context, more contextual and openly value-laden, less oriented to mastery over natural and social processes, and more accessible to the public at large.

    Notes: Summary From meteorological IGY data for the calendar year 1958, the mean meridional eddy transport of enthalpy was evaluated for the Southern Hemisphere. Levels chosen for the study were 1000, 850, 700, 500, 400, 300, 200, 150 and 100 mb. Data from 84 Southern Hemisphere and 25 equatorial Northern Hemisphere stations were used. Yearly mean quantities related to meridional eddy enthalpy flux were computed and analyzed. It was found that around 40° S there is a double-maximum zone of poleward, meridional, transient eddy enthalpy flux, the stronger transport occurring at 850 mb, and the weaker near 200 mb. The countergradient transient eddy flux regions in the low latitude mid-troposphere and in the middle and upper latitude lower stratosphere, found in previous Northern Hemisphere investigations, were observed to exist in the Southern Hemisphere also. The standing eddy heat transport, as expected, was very weak except at high latitudes where Antarctic continentality effected a large double-maximum poleward flux centered near the surface and in the lower stratosphere. The total vertically integrated enthalpy transport by the eddies was found to be poleward everywhere, reaching a maximum between 35° and 40° S.

    Notes: Abstract The NADPH oxidase complex catalyzes the formation of superoxide (O2 −) in phagocytic leukocytes. This paper reviews recent advances in our understanding of this enzyme system. Recent studies have defined conditions for reconstitution of this enzymatic activity with purified proteins in a cell-free system. The role of the individual proteins that make up the active complex, their regulation and the effects of mutations in these proteins are discussed. While these studies represent major achievements, it is clear from cytochemical investigations that additional levels of complexity exist in the modulation of the NADPH oxidase complex in vivo. A major role for cytochemical analysis in understanding the cell biological aspects of the generation of reactive oxygen species is discussed.

    Notes: Abstract  Neutrophils contain a 21-kDa phosphoprotein that undergoes rapid dephosphorylation upon stimulation of these cells with the chemoattractant N-fMet-Leu-Phe (fMLP), activators of protein kinase C [e.g., 4β-phorbol 12-myristate 13-acetate (PMA)] or the calcium ionophore A23187. This phosphoprotein was identified as the non-muscle form of cofilin by peptide sequencing and immunoblotting with specific antibodies. Evidence is presented that in neutrophils cofilin is regulated by a continual cycle of phosphorylation and dephosphorylation, and that the phosphatase undergoes activation during cell stimulation. Experiments with a wide variety of antagonists further suggested that the protein kinase that participates in these reactions may be a novel enzyme. The kinetics of cofilin dephosphorylation in neutrophils stimulated with fMLP or PMA were very similar to those observed for superoxide (O2 –) release. Immunofluorescent studies revealed that cofilin was present thouroughout the cytosol of resting neutrophils and underwent rapid translocation to the F-actin-rich, ruffled membranes of stimulated cells. Cytochemical analysis further revealed that the ruffled membranes also contained large amounts of hydrogen peroxide (H2O2), a product of the O2 –/H2O2-generating activity of stimulated neutrophils (NADPH oxidase). Cofilin is therefore well placed to participate in the continual polymerization and depolymerization of F-actin that is thought to give rise to the oscillatory pattern of H2O2 production observed under certain conditions.

Monensin and brefeldin A (BFA), inhibitors of Golgi-mediated protein secretion, rapidly perturb the transport catalytic activity of specific plasma membrane-associated efflux carriers for indole-3-acetic acid (IAA) and inhibit polar transport of IAA. To determine if these responses result solely from perturbation of the efflux carrier or whether specific auxin uptake carrier function is also affected, the influence of BFA on the cellular transport of a range of auxins with contrasting affinities for specific auxin uptake and efflux carriers was investigated in zucchini (Cucurbita pepo L.) hypocotyl tissue. In-flight addition of BFA (3 · 10−5 mol · dm−3) caused a rapid (lag 〈 10 min) and substantial (fourfold) increase in the rate of [1-14C]IAA net uptake by zucchini hypocotyl tissue. In the presence of the specific auxin efflux carrier inhibitor N-1-naphthylphthalamic acid (NPA; 3 · 10−6 mol · dm−3), BFA slightly reduced the rate of [1-14C]IAA net uptake. Stimulation of [1-14C]IAA net uptake by BFA was concentration-dependent. In the absence of BFA, net uptake of [1-14C]IAA exhibited the characteristic biphasic response to increasing concentrations of competing cold IAA but in the presence of BFA, [1-14C]IAA uptake decreased smoothly with increase in concentration of competing unlabelled IAA, indicating a loss of auxin efflux carrier activity but retention of functional uptake carriers. The half-time for mediated efflux of [1-14C]IAA from preloaded zucchini tissue was substantially increased by BFA (t1/2 = 51 min, controls; 107 min, BFA-treated). Treatment with BFA and/or NPA did not significantly affect the net uptake by, or efflux from, zucchini tissue of [1-14C]2,4-dichlorophenoxyacetic acid ([1-14C]2,4-D), a substrate for the auxin uptake carrier but not the auxin efflux carrier. Uptake of [1-14C]2,4-D declined smoothly with increasing concentrations of competing unlabelled IAA whether or not BFA was included in the uptake medium, confirming the failure of BFA to perturb auxin uptake carrier function. Transport of 1-[4-3H]naphthaleneacetic acid (1-NAA) exhibited little response to BFA or NPA, confirming that it is only a weakly transported substrate for the efflux carrier in zucchini cells.

    Notes: Abstract The growth of the anaerobic acetogenic bacterium Acetobacterium woodii DSM 1030 was investigated in fructose-limited chemostat cultures. A defined medium was developed which contained fructose, mineral salts, cysteine · HCl and Ca pantothenate (1 mg · 1−1) supplied in a vitamin supplement. Growth at high dilution rates was dependent on the presence of CO2 in the gas phase. The μ max was found to be 0.16 h−1 and the fructose maintenance requirement was 0.1 to 0.13 mmol fructose · (g dry wt)−1 · h−1. A growth yield of 61 g dry wt · (mol fructose)−1, corrected for the cell maintenance requirement and for incorporation of fructose carbon into cell biomass, was determined from the fructose consumption. A corresponding growth yield of 69 g dry wt · (mol fructose)−1 was calculated from the acetate production assuming that fructose fermentation was homoacetogenic. A YATP of 12.2 to 13.8 g dry wt · (mol ATP)−1 was calculated from these growth yields using a value of 5 mol ATP · (mol fructose)−1 as an estimate of the amount of ATP synthesised from fructose fermentation. The addition of yeast extract (0.5 g · 1−1) to the medium did not influence the μ max or cell yield. After prolonged growth under fructose-limited conditions the requirement of the culture for CO2 in the gas phase was reduced.
