    Notes: The interspecific variation in the positional and mesenteric relationship of the mammalian oviduct to the ovary observed among 41 species is summarized by distinguishing eight morphological types of relationship. The recognition of types is based on wide species differences in: the position of the oviduct with reference to the mesosalpinxal fold and ovary; the extent to which oviducal mesenteries enclose the ovary in a periovarial sac; the degree of closure and method of formation of the peritoneal opening to the periovarial sac; and the morphology of the oviduct.

    Notes: The modeling of pressure filtration of flocculated suspensions using compressional rheology and a knowledge of compressional yield stress Py(φ) and a hydraulic resistance factor r(φ)(φ is the local volume fraction of solids) is shown to yield an initial solids loading that maximizes the throughput of the filter. The optimal initial height h0 is such that the filtration time to reach a specified average volume fraction as output equals the handling time for the filter press. The maximum throughput of the press is then examined as a function of the remaining control parameters, the initial solids volume fraction φ0, and the applied piston pressure δP. The dependence of filtration time on φ0/∞00 (where φ∞is the volume fraction of solids at infinite time under applied pressure δP) enables the construction of a simple numerical model for the pressure filtration process, which accurately approximates predictions of the full compressional rheology model.

    Notes: Fermentation of xylose by Clostridium thermosaccharolyticum was studied in batch and continuous culture in which the limiting nutrient was either xylose, phosphate, or ammonia. Transient results obtained in continuous cultures with batch grown inoculum and progressively higher feed substrate concentrations exhibited ethanol selectivities (moles ethanol/moles other products) in excess of 11. The hypothesis that this high ethanol selectivity was a general response to mineral nutrient limitation was tested but could not be supported. Growth and substrate consumption were related by the equation qs(1 - Yxc)GATP = (μ/YATPmax) + m, with qs the specific rate of xylose consumption (moles xylose/hour · g cells), Yxc the carbon based cell yield (g cell carbon/g substrate carbon), GATP the ATP gain (moles ATP produces/mol substrate catabolized), μ the specific growth rate (1/h), YATPmax the ATP-based cell yield (g cells/mol ATP), and m the maintenance coefficient (moles ATP/hour · g cells). YATPmax was found to be 11.6 g cells/mol ATP, and m 9.3 mol ATP/hour · g cells for growth on defined medium. Different responses to nutrient limitation were observed depending on the mode of cultivation. Batch and immobilized cell continuous cultures decreased GATP by initiating production of the secondary metabolites, propanediol, and in some cases, D-lactate; in addition, batch cultures increased the fractional allocation of ATP to maintenance and/or wastage. Nitrogen-limited continuous free-cell cultures maintained a constant cell yield, whereas phosphate-limited continuous free-cell cultures did not. In the case of phosphate limitation, the decreased ATP demand associated with the lowered cell yield was accompanied by an increased rate of ATP consumption for maintenance and/or wastage. Neither nitrogen or phosphorus-limited continuous free-cell cultures exhibited an altered GATP in response to mineral nutrient limitation, and neither produced secondary metabolites. © 1993 John Wiley & Sons, Inc.

    Notes: Adsorption of Avicel-hydrolyzing activity was examined with respect to: mixed hardwood flour pretreated with 1% sulfuric acid for 9 s at 220°C (PTW220), lignin prepared from PTW220 by either acid or enzymatic hydrolysis, and Avicel. Experiments were conducted at 60°C for all materials, and also at 25°C for PTW220. Based on transient adsorption results and reaction rates, times were selected at which to characterize adsorption at 60°C as follows: PTW220, 1 min; lignin, 30 min; and Avicel, 45 min. Similar results were obtained for adsorption of cellulase activity to PTW220 at 25 and 60°C, and for lignin prepared by enzymatic and acid hydrolysis. For all materials, adsorption was described well by a Langmuir equation, although the reversibility of adsorption was not investigated. Langmuir affinity constants (L/g) were: PTW220, 109; lignin, 17.9; Avicel, 4.3; cellulose from PTW220, ≥187. Langmuir capacity constants were 760 for PTW220 and 42 for Avicel; the cellulase binding capacity of lignin appeared to be very high under the conditions examined, and could not be determined. At low and moderate cellulase loadings at least, the majority of cellulase activity adsorbed to PTW220 is bound to the cellulosic component. The results indicate that PTW220, and its cellulose component in particular, differ radically from Avicel with respect to adsorption. Avicel-hydrolyzing activity and CMC-hydrolyzing activities were found to bind to Avicel with a constant ratio of essentially one, consistent with adsorption of a multi-activity complex. © 1993 John Wiley & Sons, Inc.

    Notes: From the development of a compressional rheology of a flocculated suspension, a number of separation processes have been modeled. With respect to the two-stage pressure filtration process of compact formation and then consolidation, the rheological formulation appears at first examination to eb significantly at variance with the conventional engineering approach. The present model may be reconciled to the early engineering model. Moreover, the filtration parameters extracted from experiments can be related to the more fundamental rheological parameters, r(φ), the hindered settling factor, which is a function of solids volume fraction φ  -  it takes into account hydrodynamic interactions between local particles, which increase the drag forces. Useful estimates of the compact bed formation resistance and the formation time are provided as a function of applied pressure. The initial time dependence of the consolidation ratio on the elapsed consolidation time is not found to be a square root behaviour as suggested by the conventional modeling.

    Notes: Three models of a catalyst particle are examined for existence of multiple steady states depending upon whether intraparticle heat conduction or diffusion is important. The particle with constant temperature but with intraparticle diffusion is examined in detail and a necessary and sufficient condition for the stability of the steady state is obtained. Calculations are made for a particle with both intraparticle diffusion and conduction and it is shown that there may be multiple steady states some of which are unstable to small perturbations.

    Notes: Under anaerobic, carbon limited conditions, celluloytic fermentative microorganisms face a metabolic choice with respect to the allocation of relatively scarce ATP: to invest it in cells or in hydrolytic enzymes. A model is proposed that defines an allocation parameter reflecting the fractional expenditure of ATP on cell synthesis relative to the total ATP available (gross ATP synthesized less maintenance). This parameter is then incorporated into an ATP-centered model of anaerobic cellulose fermentation based on the ethanol fermentation of yeast and the cellulase system of Trichoderma reesei. Results indicate that high processing rates are possible via a consolidated bioprocessing strategy, especially at high cellulase specific activities, and that cell/cellulase allocation represents an interesting system in which to study, and perhaps exploit, microbial evolution and metabolic control. © 1998 John Wiley & Sons, Inc.     Notes: Early failures of bovine pericardial heart valves have been due to leaflet perforation/tearing and calcification. Since glutaraldehyde fixation has been shown to produce marked changes in leaflet mechanics and has been linked to the development of calcification, alternative crosslinking techniques have been suggested as means to overcome these obstacles. We have examined the low strain rate viscoelastic behavior of bovine pericardium: (1) fresh; (2) chemically treated with glutaraldehyde, cyanimide, or polyglycidyl ether; or (3) physically treated by freeze-drying or heat-drying. Shrinkage temperature tests were conducted to assess intrahelical crosslinking. Polyglycidyl ether and glutaraldehyde both produced substantial crosslinking, with the shrinkage temperature rising above 80°C. Mechanical changes were nearly equivalent, both showing decreased stress relaxation and increased extensibility consistent with intrahelical crosslinking and shrinkage during fixation. Cyanimide, known to crosslink pure collagen materials, showed no evidence of crosslinking intact tissue. Heat-drying, also effective in pure collagen preparations, produced an increase in UTS and tissue modulus, but otherwise left the tissue unchanged. Freeze-drying had no mechanical effect, and therefore provides an attractive means for the storage of connective tissues for later mechanical testing.
