Components of the Green Deathcap Toadstool, Amanita phalloides, LI. - The Cycloamanides, Monocyclic Peptides; Isolation and Elucidation of Structure of a Cyclic Heptapeptide (CyA B) and of Two Cyclic Octapeptides (CyA C and CyA D)Apart from the known compounds antamanide (5) and phallin A (1), the new cyclic peptides cyclo-(Pro-Ser-Phe-Phe-Phe-Pro-Ile) (2), cyclo-(Pro-Met(O)-Leu-Gly-Phe-Leu-Val-Leu) (3, R- and S-sulfoxides), and cyclo-(Pro-Met(O)-Leu-Gly-Phe-Leu-Pro-Leu) (4, R- and S-sulfoxides) have been isolated from the fraction of the components of Amanita phalloides which is soluble in ethyl acetate but not in petroleum ether. Their structures were established by the dansyl modification of Edman degradation of suitable linear peptides. The monocyclic peptides (except antamanide) are designated as cycloamanides (CyA), 1 now being named CyA A, 2 CyA B, 3 CyA C and 4 CyA D.

Aus dem in Ethylacetat, nicht aber in Petrolether löslichen Anteil der Inhaltsstoffe von Amanita phalloides, aus dem seinerzeit Antamanid (5) isoliert wurde, wurden durch eine Reihe chromatographischer Verfahren außer dem bekannten Phallin A (1) die weiteren monocyclischen Peptide cyclo-(Pro-Ser-Phe-Phe-Phe-Pro-Ile) (2), cyclo-(Pro-Met(O)-Leu-Gly-Phe-Leu-Val-Leu) (3, R- und S-Sulfoxide) und cyclo-(Pro-Met(O)-Leu-Gly-Phe-Leu-Pro-Leu) (4, R- und S-Sulfoxide) isoliert und in ihrer Konstitution durch Dansyl-Edman-Abbau geeigneter linearer Peptide aufgeklärt. Die monocyclischen Peptide (außer Antamanid) werden als Cycloamanide (CyA) bezeichnet, wobei 1 die Bezeichnung CyA A, 2 CyA B, 3 CyA C und 4 CyA D erhalten.
