    Notes: Abstract We have identified and obtained the full-length clone of RREBP49, a human nuclear factor which specifically interacts with the Rev-responsive element (RRE) sequence of human immunodeficiency virus type 1. Sequence analysis revealed that RREBP49 is highly homologous to hnRNP F protein and contains three repeated RNA-binding domains. Binding assays demonstrated that Rev and RREBP49 bind to different subregions on the RRE sequence and that binding is mutually nonexclusive. Blocking of endogenous RREBP49 expression by an antisense construct increases Rev activity in CV-1 cells, indicating that RREBP49 and Rev may play antagonistic roles in HIV-1 replication. RREBP49 may function as a splicing factor or a nuclear retention factor for unspliced mRNAs. However, only a slight decrease of Rev activity was observed when exogenous RREBP49 was introduced into CV-1 cells by pSVL-RREBP49 expression vector. This may be explained by a high endogenous level of RREBP49 which is above optimal. Alternatively, additional cellular factors may be required for RREBP49-mediated inhibition of Rev.

    Notes: Abstract The initial step in the signaling cascade of the growth factor activin involves its binding to the extracellular domain of the activin type II receptor. This receptor domain contains 10 cysteine residues which are engaged in intramolecular disulfide bonds. To elucidate the structural framework of this domain we have characterized its disulfide-bonding pattern using an extracellular fragment of the receptor which binds activin A with high affinity. By combining proteolysis with mass spectroscopy and chemical sequence analysis, the disulfide connectivity was determined to be as follows: C1–C3, C2–C4, C5–C8, C6–C7, and C9–C10. A similar disulfide arrangement occurs in a family of snake toxins for which the three-dimensional structure is known.

    Notes: Abstract A manual Edman degradation protocol has been developed that allows the identification of phosphorylation sites in 32P-labeled peptides at the subpicomole level. By using both a volatile reagent, trifluoroethyl isothiocyanate, and volatile buffers, extraction steps are rendered unnecessary and cycle times can be reduced to 45 min. The protocol was employed to identify the site of phosphorylation in phosphoserine- and phosphotyrosine-containing peptides.

    Notes: Abstract The Australian Federal Government introduced in 1994 the Trade Practices Amendment (Origin Labelling) Bill in the House of Representatives, Parliament of Australia, to establish a scheme to govern representations about the origin of consumer goods. This amendment to the Trade Practices Act (1974) applies only to Australian products, and provides two labelling alternatives, that is Product of Australia (no imported components and processing has been done completely in Australia) and Made in Australia (with some imported components). The Origin Labelling Bill 1994 is currently pending in the Senate, Parliament of Australia, for various reasons. Apart from the investigation into legal matters, a major aim of the research to be reported here was to question the impact of country of origin on the consumers' buying decision process, and in particular the impact of the Australian Made logo on sales of consumer goods. This logo is provided by the Advance Australia Foundation as a part of the Buy Australian Made campaign, started in 1986 to reduce the Australian import penetration. A Townsville-based consumer survey, conducted as a part of the research, showed that price, quality, and value for money were the over-riding factors in purchasing decisions. The survey sampled 647 interviewed consumers, and included an experiment in which business shirts and tea bags were used to determine reactions to country of manufacture information. It was found, for example, that the addition of a logo to a shirt had no positive influence on the Australian shirt's propensity to be selected as the preferred shirt when compared to the imported shirts.

    Notes: The Stoichiometry in the Determination of Water Using Karl Fischer ReagentThe stoichiometry of the Karl Fischer reaction is not unambiguous. The titer of a Karl Fischer solution rises with the amount of water, because sulphur trioxide which is formed as an intermediate consumes water too. An 1:1-stoichiometry (H2O:I2) can be simulated by alcohols, because these solvents also react with iodine and sulphur dioxide leading to additional consumption of iodine during the titration. This interpretation of the results of Karl Fischer titrations is supported by the isolation or identification of pertinent reaction products.

    Notes: Rheo-optical Fourier transform infrared spectroscopy was used to analyze the orientational behavior of hard and soft segments of thermoplastic poly(ether urethane)s (TPU-Et) and poly(ester urethane)s (TPU-Es) and their blends with a 20 mass-% contribution of either polyethylene (PE) or polypropylene with an amorphous rubber phase (PP). The blends show a reduced stress level, lower elongation-to-break and a lower degree of orientation of the segments  -  especially for the TPU-Es blends  -  compared to those of pure TPU's when uniaxially elongated. Blends of TPU-Et with PP show reduced stress but increased strain compared to pure films of TPU-Et. This is caused by an improved adhesion of the rubber phase in the polypropylene and the polyether phase of TPU-Et. Optical micrographs visualize deformed polyolefin particles in elongated films of TPU-Et/PP blends. In the TPU-Et/PP blend we could achieve equivalent orientation for the polyolefin phase and the segments of TPU-Et caused by this higher phase adhesion compared to the other blends. Orientation functions (OF's) of absorption bands which specifically represent hard or soft polyurethane segments or which are characteristic of the polyolefin phase have been calculated to monitor the molecular alignment due to the mechanical treatment.

    Notes: The Decomposition of Acidic Karl Fischer Reagent in MethanolThe reaction between sulfur dioxide and iodine in methanol is started by traces of water in the solvent. Hydrogen iodide is formed and reacts with methanol to produce more water until all iodine is used up. An addition compound between iodine and hydrogen sulfite was found as an intermediate and characterized by Raman spectroscopy. Elementary sulfur is formed in a second reaction.
