    Description: Tumour suppressor p53, which is encoded by the TP53 gene, is widely known to play an important role in response to DNA damage and various stresses. It has recently been reported that p53 regulates glucose metabolism and that an increase in p53 protein level is induced after serum deprivation or treatments with a natural compound, trans -Resveratrol (Rsv). In this study, we constructed a Luciferase expression vector, pGL4-TP53-551, containing 551 bp of the 5’-upstream region of the human TP53 gene, which was then transfected into HeLa S3 cells. A Luciferase assay showed that Rsv treatment increased the promoter activity of the TP53 gene in comparison to that of PIF1 . Detailed deletion and mutation analyses revealed that Nkx-2.5 and E2F-binding elements are required in addition to duplicated GGAA (TTCC), for the regulation of TP53 promoter activity. In this study, it is suggested that the transient induction of TP53 gene expression by Rsv treatment might be partly involved in its anti-aging effect through maintenance of chromosomal DNAs.

    Description: Frequency-domain loop–loop electromagnetic (EM) methods are sensitive to the magnetic susceptibility of the Earth as well as its resistivity. Thus, inversion techniques have been used to simultaneously reconstruct both resistivity and susceptibility models from EM data. However, to take full advantage of inversion methods, calibration errors must be assessed and removed because ignoring them can result in misleading models. We present a multidimensional inversion method that jointly inverts EM and direct current (DC) resistivity data to derive offset errors as well as resistivity and susceptibility models, assuming that calibration errors can be represented by in-phase and quadrature offsets at each frequency. Addition of independent data such as DC data is effective for more accurately estimating the offsets, resulting in more reliable subsurface models. Synthetic examples involving small-loop EM data show that simultaneous inversion for resistivity and susceptibility is not stable, because of strong correlations between in-phase offset parameters and background susceptibility, but that the offsets are well determined when the data misfit is reduced rapidly in the early iteration step. Improvements achieved by joint inversion are mainly on the resistivity model. For airborne electromagnetic (AEM) data, the inversion process is stable, because AEM data are acquired using more loop–loop geometries and a wider range of frequencies. As a result, both the resistivity and susceptibility models are significantly improved by joint inversion.

    Description: The catalytic behavior of various noble metal nanoparticles (NPs) supported directly on multiwalled carbon nanotubes (MWCNTs) was observed using environmental transmission electron microscopy (E-TEM). Gasification of the MWCNTs via catalytic hydrogenation or oxidation progressed at ~450°C in conjunction with certain noble metal NP catalysts at the interface between MWCNTs and the NPs. During such catalytic reactions, the NPs were observed to move rapidly over the MWCNT surfaces. The mobility and wettability of the NPs varied depending on the particular metal NPs employed and the ambient atmosphere. While rhodium NPs exhibited high wettability under both hydrogen and oxygen atmospheres, the wettability of platinum, palladium and iridium NPs on MWCNTs varied with the atmosphere. The metal NPs seemed to have high degrees of crystallinity while their morphologies fluctuated throughout the catalytic reactions.

    Description: Although nitric oxide (NO) is an important signaling molecule in bacteria and higher organisms, excessive intracellular NO is highly reactive and dangerous. Therefore, living cells need strict regulation systems for cellular NO homeostasis. Recently, we discovered that Streptomyces coelicolor A3(2) retains the nitrogen oxide cycle (NO 3 – -〉NO 2 – -〉NO-〉NO 3 – ) and nitrite removal system. The nitrogen oxide cycle regulates cellular NO levels, thereby controlling secondary metabolism initiation (red-pigmented antibiotic, RED production) and morphological differentiation. Nitrite induces gene expression in neighboring cells, suggesting another role for this cycle as a producer of transmittable intercellular communication molecules. Here, we demonstrated that ammonium-producing nitrite reductase (NirBD) is involved in regulating NO homeostasis in S. coelicolor A3(2). NirBD was constitutively produced in culture independently of GlnR, a known transcriptional factor. NirBD cleared the accumulated nitrite from the medium. Nir deletion mutants showed increased NO-dependent gene expression at later culture stages, whereas the wild-type M145 showed decreased expression, suggesting that high NO concentration was maintained in the mutant. Moreover, the nir deletion mutant produced more RED than that produced by the wild-type M145. These results suggest that NO 2 – removal by NirBD is important to regulate NO homeostasis and to complete NO signaling in S. coelicolor .
