    Description / Table of Contents: Sustainability in an Imaginary World explores the social agency of art and its connection to complex issues of sustainability. Over the past decade, interest in art's agency has ballooned as an increasing number of fields turn to the arts with ever-expanding expectations. Yet just as art is being heralded as a magic bullet of social change, research is beginning to throw cautionary light on such enthusiasm, challenging the linear, prescriptive, instrumental expectations such transdisciplinary interactions often imply. In this, art finds itself at a treacherous crossroads, unable to turn a deaf ear to calls for help from an increasing number of ostensibly non-aesthetic fields, yet in answering such prescriptive urgencies, jeopardizing the very power for which its help was sought in the first place. This book goes in search of a way forward, proposing a theory of art aiming to preserve the integrity of arts practices within transdisciplinary mandates. This approach is then explored through a series of case studies developed in collaboration with some of Canada's most prominent artists, including internationally renowned nature poet Don McKay; Italian composer and Head of Vancouver New Music, Giorgio Magnanesi; the renowned Electric Company Theatre, led by Kevin Kerr; and finally through a largescale multimedia installation aiming to reimagine the relationship between climate, culture, and human agency. Sustainability in an Imaginary World will be of great interest to students and scholars of arts-based research fields, sustainability studies, and environmental humanities

    Notes: The initiation of many physiologically important biochemical reactions is dependent on the binding of a molecule or molecules from solution to an appropriate solid surface such as a cell membrane. The quantitative study of dynamic interactions of molecules in solution with immobilized surfaces under physiologically relevant conditions presents a significant physical problem. Traditionally these measurements have been performed by discontinuous, nonequilibrium assays that cannot accurately model in vivo reaction mechanisms. Total internal reflection fluorescence spectroscopy (TIRFS) is an analytical technique that is based on the excitation of fluorescence by means of an exponentially decaying surface energy wave called the evanescent wave. The limited "penetration depth'' of the evanescent wave leads to excitation of fluorescent molecules at or near the surface. This phenomena makes TIRFS uniquely qualified for the dynamic fluorescence study of liquid-solid interface binding reactions. The instrument described here permits the powerful technique of TIRFS to be used by biomedical researchers in their investigation of biochemical interface reactions. The fully automated, menu driven instrument allows the real time dynamic measurement of binding of fluorescent-labeled molecules in solution at physiologically relevant concentrations to appropriate immobilized surfaces.The novel design of the total internal reflection element enables the study of binding from essentially opaque and highly scattering solutions such as whole blood. Measurements may be performed under static, stirred, or flow conditions. The presently described TIRFS instrument is capable of tracking a binding reaction with evanescent fields established by two identical reflecting light beams with different reflection angles. This instrumental feature allows the dynamic measurement not only of the primary binding event but also of fluctuations in the distance between the optical interface and the point of signal generation. Data from experiments involving fluorescently labeled molecules binding to cell monolayers as well as binding experiments performed in whole blood are shown to verify the instrument performance. Due to its extraordinary capabilities and its user friendly design, the developed TIRFS instrument opens up a large number of relevant but previously unmeasurable experimental systems for investigation.

    Notes: Sensory ratings, respiratory behavior and eye blink rate were recorded in 11 nonsmokers exposed to a no-smoking (CONTROL) condition or to environmental tobacco smoke (ETS) from one of three kinds of cigarettes: a Kentucky reference (1R4F), an ultra-low tar (ULT) and one that heats tobacco (TEST).For each two-hour session in a controlled-environment room, two smokers “puffed” unlit cigarettes, for the CONTROL condition, or smoked 16 cigarettes to generate ETS (sidestream plus exhaled mainstream smoke). Concentrations of nicotine and respirable suspended particles (RSP) observed with the 1R4F and ULT cigarettes were 10- to 20-fold higher than those typically found in field sampling studies of office and restaurant smoking environments. The ULT and TEST cigarettes resulted in ETS analyte concentrations that were 77-100% and O-56%, respectively, of those observed with 1R4E Sensory ratings (odor, irritation, annoyance, acceptability) with the two tobacco-burning cigarettes were similar and were much greater than those recorded for the TEST or CONTROL conditions which were, in turn, rated similarly. Respiratory parameters (respiratory rate, tidal volume, expiratory time), which generally failed to discriminate the TEST and CONTROL conditions, were altered during exposure to ETS from the 1R4F and, to a lesser extent, the ULT cigarette. Eye blink rate was not significantly altered by ETS from any of the cigarettes.

    Notes: Abstract Cell-free synovial fluid from patients with rheumatoid arthritis contains soluble and insoluble IgG-containing immune complexes which activate reactive oxidant production in human neutrophils. In this report we have measured the effects of inhibitors of signal transduction pathways on neutrophil activation by these complexes and also following activation by synthetic soluble and insoluble immune complexes made from human serum albumin (HSA) and anti-(HSA) antibodies. In all aspects studied, the soluble rheumatoid complexes and the soluble synthetic complexes were indistinguishable in the ways in which they activated neutrophils. Activation of reactive oxidant production in response to these soluble complexes was completely inhibited by pertussis toxin (indicating G-protein coupling of receptor occupancy), completely insensitive to staurosporine (indicating that oxidant production did not require protein kinase C activity), only marginally (〈30%) inhibited by butanol (indicating that dependence upon activity of phospholipase D was minimal), and completely inhibited by chloracysine, an inhibitor of phospholipase A2. In contrast, activation of reactive oxidant production in response to the insoluble rheumatoid or insoluble synthetic immune complexes was largely pertussis toxin insensitive, inhibited by 〉50% by staurosporine, inhibited by 〉50% by butanol, and completely inhibited by chloracysine. These results show that the receptor-mediated signal transduction systems activated by the soluble and insoluble immune complexes are different. Because the soluble complexes activate a transient burst of reactive oxidant secretion from primed neutrophils, the mechanisms regulating either the release or the intracellular production of oxidants within rheumatoid joints are distinct and hence may be pharmacologically modified independently of each other.

    Notes: Abstract Stimulation of human neutrophils with the chemotactic peptide fMet-Leu-Phe results in activation of a rapid, transient burst of oxidant secretion, which reaches a maximal rate by about 1 min after stimulation. This phase of oxidant secretion is then followed by intracellular oxidant production, which is detected by luminol chemiluminescence but not by assays such as cytochrome c reduction or scopoletin oxidation. The rapid phase of oxidant secretion requires increases in intracellular free Ca2+ and phospholipase A2 activity, but not the activities of phospholipase D or D or protein kinase C. In contrast, intracellular oxidant production requires the activities of phospholipase D and protein kinase C. A model is thus proposed suggesting the sequential activation of different phospholipases which activate oxidase molecules on the plasma membrane or else from the membranes of specific granules.

    Notes: Abstract This article examines two examples of what has been called mandated science - risk analysis and long-term energy demand forecasting - with a view to disentangling the implicit assumptions underlying their use in public policy debates and in the policy process. In both fields, there has emerged a growing and very similar challenge to traditional views of the role and status of mandated science in informing decision-making. This challenge has led to the reformulation of some of the attributes of the standard view, without however causing changes in the underlying view of the nature of the problem or the role of science in principle. A deeper critique suggests that mandated science is inherently value-laden, producing conclusions that are strongly affected by theoretical presuppositions that in turn reflect underlying values (e.g., about rationality, knowledge or the role of technology). Thus a sharp separation of science and policy is untenable and amounts to a hidden policy bias in favor of the business-as-usual policies supported by conventional science. There is a need for a new concept of science, one that is more integrated into the policy context, more contextual and openly value-laden, less oriented to mastery over natural and social processes, and more accessible to the public at large.
