    Notes: Summary The nucleus lateralis tuberis (NLT) system of the teleost fish Gillichthys mirabilis was studied with respect to the anatomical organization and distribution of the neurosecretory neurons and the nature of their secretory material. NLT neurons occur in several different areas in the tuberal region of the hypothalamus, located immediately above the pituitary gland. Only the lateral and rostral NLT neurons show definite secretory activity. These neurons, generally of large size, contain numerous large granulated vesicles (LGV), 900–1,000 Å in diameter; on the other hand, the medial and ventrolateral neurons, forming a single layer of cells, are of small size and do not appear to be secretory. LGV are positive after ethanol-phosphotungstic acid (E-PTA) and zinc iodide-osmium tetroxide (ZIO) impregnations. Lateral and rostral neurosecretory neurons also show strong yellow-to-green specific fluorescence after the Falck-Hillarp technique, a strong indication that LGV are the site of catecholamine storage.

    Notes: Summary The neurohypophysis of the South American lungfish Lepidosiren paradoxa has been studied with light and electron microscopy, including the Falck-Hillarp technique for catecholamines. The pars nervosa hypophyseos is a well-marked, dorsally located subdivision of the pituitary gland composed of lobes or follicles, each one constituted of a central core of ependymal cells, a subependymal hilar region made up of nerve fibers and a peripheric palisade zone of nerve endings which contact capillary vessels. Four types of neurosecretory axons can be distinguished under the electron microscope. Type I, the most common, contains spherical elementary granules of high electron density, 1500–1800 Å in diameter. The scarce type II axons contain irregularly-shaped elementary granules. Type III contains only small clear vesicles, 400–600 Å in diameter. Type IV, mostly present in regions of the gland contacting the pars intermedia, contain large granulated vesicles, 900–1000 Å in diameter. The Falck-Hillarp technique revealed axons with a positive reaction for catecholamines at sites corresponding approximately to the location of type IV of the electron microscope. Ependymal cells are of large size, linking the cerebrospinal fluid, the nerve endings and the blood vessels. A conspicuous membrane-bound, spherical dense material, 1400–2000 Å in diameter, is observed in both the apical and vascular processes of these cells. The ependymal processes which traverse the hilar and palisade regions contain structures resembling degenerated neurosecretory axons. These results are discussed in relation with the currently available information on the comparative anatomy of the pars nervosa. The possible functional significance of ependymal cells and of each type of axon are also discussed.     Notes: Summary The action of luteinizing releasing hormone (LH-RH) on FSH and LH-gonadotrophs from the anterior pituitary glands of normal rats has been studied with electron microscopy. After 15 min. incubation with LH-RH, LH cells showed secretory granules undergoing exocytosis, but FSH did not appear to release secretory products. On the other hand, incubations with the releasing hormone for 3 and 6 hr stimulated exocytosis from both FSH and LH-gonadotrophs. Of more interest was the finding that FSH cells appeared as typical “signet cells” of castration when glands were incubated for 3 and 6 hr with the releasing hormone. The appearance of such “signet cells” was prevented if puromycin was added to the medium together with LH-RH.

    Notes: Summary The rostral pars distalis (RPD) of the teleost Mugil platanus from animals pretreated with reserpine or 6-hydroxydopamine (6-HODA) were assayed for dopamine (DA) or noradrenaline (NA) or for prolactin hormone. Such determinations were coupled with electron microscopy. It was found that reserpine and 6-HODA produced a significant decrease in the content of DA, NA, and prolactin. Electron microscope studies revealed that prolactin cells became activated as judged by ultrastructural criteria. After 6-HODA treatment type “B” neurosecretory fibers entering the RPD became selectively destroyed. These observations lead us to suggest that prolactin secretion is under inhibitory control by type “B” neurosecretory fibers of adrenergic nature.

    Notes: Summary The innervation of the pituitary gland of the teleost fish Gillichthys mirabilis was studied with light and electron microscopy in order to determine its nature and distribution. Two types of neurosecretory fibers (“A” and “B”) are present in the adenohypophysis. Type “A” fibers containing elementary neurosecretory granules, 1,500–1,600 Å in diameter, perforate the basement membrane in the neurointermediate lobe and make synaptoid contacts with MSH cells. Type “B” fibers containing large granulated vesicles (LGV), 900–1,000 Å in diameter, perforate the basement membrane in all lobes of the pituitary gland and terminate in direct contact with the several adenohypophysial cell types. At the electron-microscope level, LGV from type “B” fibers show a positive reaction to zinc iodide-osmium tetroxide (ZIO) impregnation. Reserpine treatment resulted in a depletion of their dense cores if osmium tetroxide was used as the only fixative, whereas double fixation with aldehydes and osmium tetroxide did not reveal appreciable changes. Yellow-to-green fluorescent fibers were detected in the pituitary gland after the Falck-Hillarp technique at sites corresponding approximately to the location of type “B” fibers, strongly suggesting the monoaminergic nature of the latter. After hypophysectomy, medial and lateral neurons of the nucleus lateralis tuberis (NLT) undergo retrograde degeneration. This finding, together with the morphological and cytochemical similarities of the LGV in NLT neurons and those in the type “B” fibers, suggests that the fibers originate from certain NLT neurosecretory neurons.

    Notes: Summary Five categories of granulated cells were distinguished by their ultrastructural features, and quantitative analyses were made of the pars distalis cells in normal and castrated lizards. The gonadotropin-producing cell was identified on the basis of its uniform distribution in the gland as well as from cytological changes resulting from castration. The secretory granules of the gonadotropic cell vary in size (100–500 mμ) and density, and lipid bodies are commonly present. Following castration, the endoplasmic reticulum proliferates, forming many small, rough-surfaced, dilated cisternae which do not coalesce greatly as in other vertebrate species. Degranulation is accompanied by hypertrophy and hyperplasia of the mitochondria and by the appearance in the cytoplasm of conspicuous clusters of microfilaments. The designated gonadotropic cell was the only class of secretory cell showing consistent changes following three weeks of castration. In addition to the uniformly distributed gonadotrope cell, two secretory cells occur mainly in the rostral half of the gland, and two in the caudal half. Tentative identification of the cell types is discussed in the light of available information on the localization of the hormones in the pars distalis of this species.

    Notes: Summary Seven days of dehydration produce alterations in the Purkinje cells of the toad, Bufo arenarum Hensel. Aldehyde-fuchsin-positive granules appear in the cytoplasm of the cells and at the same time modifications take place in cytoplasmatic and nuclear ribonucleoproteins. These modifications consist of an increase in nucleolar volume and appearance of a nuclear cap, polarized towards the granules.

    Notes: Summary The hypothalamic neurosecretory system of normal dogs was studied by light and electron microscopy after perfusion-fixation. In the supraoptic nucleus most neurons are loaded with elementary neurosecretory granules having a content of low electron density. Neurons with less neurosecretory material and signs of enhanced synthetic activity, as recognized by the changes in the endoplasmic reticulum, were also observed. The vesiculated neurons ofJewell were studied under the electron microscope and various stages of development were described. It was postulated that they originate by a localized process of cytoplasmic cytolysis which ends in the formation of a large aqueous intracellular cavity limited by a plasma membrane. The possible significance of these vesiculated neurones is discussed. Some few myelinated neurosecretory axons are found in the supraoptic nucleus. The neurons of the paraventricular nucleus are smaller and contain less neurosecretory material. This is abundant and very pale in the axons. The median eminence consists of an inner zone, mainly occupied by the neurosecretory axons of the hypothalamic-neurohypophysial tracts, and an outer zone in which some neurosecretory axons end on the capillary of the portal system. This outer zone contains numerous axons with the axoplasm rich in neurofilaments and some containing granulated and non-granulated synaptic vesicles. Some neurons with granulated vesicles were observed in this region. The adrenergic nature of these neurons and axons is postulated. The infundibular process of the neurohypophysis shows small axons with discrete amounts of elementary granules and vesicles of synaptic type at the endings. Some enlarged axons having, in addition, large polymorphic bodies are observed and related to the Herring bodies. The size and morphology of the granules are analyzed along the entire hypothalamic-neurohypophysial system. The changes in diameter and electron density are related to the maturation of the granules and the possible significance of such evolution.

    Notes: Summary The arcuate complex, comprising the nucleus and the outer zone of the median eminence, was studied under the electron microscope during the whole sexual cycle of the female rat. The content of granulated vesicles in neurons and terminals was minimal at estrus, rose during diestrus and reached maximal levels during the morning of proestrus. In the afternoon of proestrus the number of granulated vesicles suddenly decreased and lysosomes seemed to become activated similar changes were found in the nerve endings of the outer zone of the median eminence. These cyclic changes observed in the arcuate complex are discussed in relation to the knowledge on the histophysiology of this region of the hypothalamus and specially to the probable regulatory effect of monoamines on the secretion of gonadotrophins.

    Notes: Summary The connective epineural sheath of the abdominal mass ganglia of the land snail Cryptomphallus aspersa was studied by light and electron microscopy. In this sheath several cellular types were found: fibroblasts, macrophage cells, a special type rich in mitochondria; elongated cells which proved to be muscle cells of paramyosin type and glial cells with dense bodies in close contact with neurosecretory axons. Special attention was paid to the filaments of the muscle cells. Thin and thick ones were found, the last with cross bands with a repeating period of 150 Å. These findings are discussed in relation with the physiological role of the epineural connective sheath in metabolic and mechanical functions.
