    Description: Phialophora as defined by its type species P. verrucosa is a genus of Chaetothyriales, and a member of the group known as ‘black yeasts and relatives’. Phialophora verrucosa has been reported from mutilating human infections such as chromoblastomycosis, disseminated phaeohyphomycosis and mycetoma, while morphologically similar fungi are rather commonly isolated from the environment. Phenotypes are insufficient for correct species identification, and molecular data have revealed significant genetic variation within the complex of species currently identified as P. verrucosa or P. americana. Multilocus analysis of 118 strains revealed the existence of five reproductively isolated species apparently having different infectious potentials. Strains of the sexual morph Capronia semiimmersa cluster within P. americana. The newly defined taxa differ markedly in their predilection for the human host.

    Description: Mean, clear-sky surface temperature of the Greenland Ice Sheet was measured for each melt season from 2000 to 2005 using Moderate-Resolution Imaging Spectroradiometer (MODIS)–derived land-surface temperature (LST) data-product maps. During the period of most-active melt, the mean, clear-sky surface temperature of the ice sheet was highest in 2002 (−8.29 ± 5.29°C) and 2005 (−8.29 ± 5.43°C), compared to a 6-year mean of −9.04 ± 5.59°C, in agreement with recent work by other investigators showing unusually extensive melt in 2002 and 2005. Surface-temperature variability shows a correspondence with the dry-snow facies of the ice sheet; a reduction in area of the dry-snow facies would indicate a more-negative mass balance. Surface-temperature variability generally increased during the study period and is most pronounced in the 2005 melt season; this is consistent with surface instability caused by air-temperature fluctuations.

    Notes: Insulin has a concentration of 10−8–10−11 M in the blood which ensures that it circulates and exerts its physiological functions in vivo as a monomer. The crystal structure of monomeric porcine desB1-B2 despentapeptide (B26–B30) insulin (DesB1-2 DPI) with Mr = 4934 Da has been determined at 1.65 Å resolution using the molecular replacement method. A structural comparison between DesB1-2 DPI and 2Zn insulin reveals that the conformation of DesB1-2 DPI is more similar to molecule I than molecule II of 2Zn insulin. The remarkable conformational difference between B25-Phe in DesB1-2 DPI and B25-Phe in despentapeptide (B26–B30) insulin (DPI) indicates that the residue B25-Phe possesses great flexibility and mobility.

    Notes: Staphylokinase, a fibrin-specific plasminogen activator, was highly expressed in Escherichia coli and purified by ion-exchange and gel-filtration chromatography. The purified recombinant staphylokinase was fully active and readily crystallized against 1.2 M sodium citrate in 100 mM Tris–HCl buffer at pH 8.0 using the hanging-drop method. Crystals of staphylokinase diffract to better than 2.2 Å resolution. The crystal belongs to the tetragonal space group P41212 or its enantiomorph with unit-cell parameters a = b = 67.5, c = 150.1 Å. There are two molecules in the asymmetric unit. In this paper, we described the first crystallization of a kind of plasminogen activator and present the results of preliminary X-ray diffraction data from the native protein.

    Notes: An electron crystallographic image-processing technique based on the combination of high-resolution electron microscopy and electron diffraction has been developed to investigate the commensurate structural modulation in the high-Tc superconductor (Pb0.5Sr0.3Cu0.2)Sr2(Ca0.6Sr0.4)Cu2Oy. After symmetry averaging, a structure image was obtained by image deconvolution at the resolution limited by that of the electron microscope. Then phase extension was employed to enhance the image resolution up to about 1.25 Å by means of the electron diffraction data corrected with an empirical method. In the final projected potential map, the occupational and/or positional modulation is clearly observed for all atoms, including oxygen. The key points of determining superstructures by the technique are studied and discussed.
