    Description: DNA methylation is critical for normal development and plays important roles in genome organization and transcriptional regulation. Although DNA methyltransferases have been identified, the factors that establish and contribute to genome-wide methylation patterns remain elusive. Here, we report a high-resolution cytosine methylation map of the murine genome modulated by Lsh, a chromatin remodeling family member that has previously been shown to regulate CpG methylation at repetitive sequences. We provide evidence that Lsh also controls genome-wide cytosine methylation at nonrepeat sequences and relate those changes to alterations in H4K4me3 modification and gene expression. Deletion of Lsh alters the allocation of cytosine methylation in chromosomal regions of 50 kb to 2 Mb and, in addition, leads to changes in the methylation profile at the 5′ end of genes. Furthermore, we demonstrate that loss of Lsh promotes—as well as prevents—cytosine methylation. Our data indicate that Lsh is an epigenetic modulator that is critical for normal distribution of cytosine methylation throughout the murine genome.

    Description: The distributed H -consensus filtering problem is investigated for a class of discrete time-varying non-linear systems on a finite horizon. The topology of the sensor networks is assumed to be Markovian switching and the missing measurements (packet dropouts) problem is also considered. Based on the recursive linear matrix inequalities, an effective distributed H -consensus filter is designed, which is suitable for online computation. A numerical example is given to verify the effectiveness of the obtained results.

    Description: Broadly, tissue regeneration is achieved in two ways: by proliferation of common differentiated cells and/or by deployment of specialized stem/progenitor cells. Which of these pathways applies is both organ- and injury-specific. Current models in the lung posit that epithelial repair can be attributed to cells expressing mature lineage markers. By contrast, here we define the regenerative role of previously uncharacterized, rare lineage-negative epithelial stem/progenitor (LNEP) cells present within normal distal lung. Quiescent LNEPs activate a DeltaNp63 (a p63 splice variant) and cytokeratin 5 remodelling program after influenza or bleomycin injury in mice. Activated cells proliferate and migrate widely to occupy heavily injured areas depleted of mature lineages, at which point they differentiate towards mature epithelium. Lineage tracing revealed scant contribution of pre-existing mature epithelial cells in such repair, whereas orthotopic transplantation of LNEPs, isolated by a definitive surface profile identified through single-cell sequencing, directly demonstrated the proliferative capacity and multipotency of this population. LNEPs require Notch signalling to activate the DeltaNp63 and cytokeratin 5 program, and subsequent Notch blockade promotes an alveolar cell fate. Persistent Notch signalling after injury led to parenchymal 'micro-honeycombing' (alveolar cysts), indicative of failed regeneration. Lungs from patients with fibrosis show analogous honeycomb cysts with evidence of hyperactive Notch signalling.     Description: We report a draft sequence for the genome of the domesticated silkworm (Bombyx mori), covering 90.9% of all known silkworm genes. Our estimated gene count is 18,510, which exceeds the 13,379 genes reported for Drosophila melanogaster.     Description: 〈span〉〈div〉ABSTRACT〈/div〉High-performance audio-frequency magnetotelluric (AMT) instrument is one means of obtaining high-quality electromagnetic (EM) data. To improve the ability of AMT system to obtain high-quality data, this paper presents a design for a high-performance analog front-end circuit for AMT instrument. It mainly consists of the input protection, preamplifier, passive high pass filter, power frequency notch filter, programmable amplifier, and active low pass filter. In addition, this paper proposes a design of low-noise, high-performance preamplifier, which improves the common-mode rejection ratio (CMRR) of analog front-end circuit and effectively enhances the signal-to-noise ratio (SNR) of the circuit. The front-end circuit utilized two-stage twin-T notch filter to effectively suppress the strong interference of fundamental component of power frequency. Also, it used signal relays to control circuit gain and selection of cutoff frequency of anti-aliasing filter, resulting in the improvement of the capability of the analog-to-digital Converter (ADC) to distinguish weak EM signal.The measured results of the electric field and magnetic field channel showed that: 1) The circuit works in frequency range of 1 Hz∼100 kHz; 2) The CMRR values of the preamplifier of electric field channel at low frequencies (1 Hz∼1 kHz) are 111 dB and 97 dB when the gains are 20 dB and 6 dB respectively; 3) The maximum attenuation fundamental power frequency can reach −39.46 dB and −39.04 dB respectively; 4) The total harmonic distortion rate at 1 kHz is 0.022% and 0.029% respectively; 5) The input noise levels of electric field channel are 12.67nV / @10Hz and 8.15V / @1kHz, while the input noise levels of magnetic field channel are 8.97nV / @10Hz and 6.16V / @1kHz; and 6) In conclusion, the analog front-end circuit is superior to meet the requirements of the AMT methods, and provides a useful reference for the development of AMT instrument. 〈/span〉
