    Description: Immune clearance and resource limitation (via red blood cell depletion) shape the peaks and troughs of malaria parasitemia, which in turn affect disease severity and transmission. Quantitatively partitioning the relative roles of these effects through time is challenging. Using data from rodent malaria, we estimated the effective propagation number, which reflects the relative importance of contrasting within-host control mechanisms through time and is sensitive to the inoculating parasite dose. Our analysis showed that the capacity of innate responses to restrict initial parasite growth saturates with parasite dose and that experimentally enhanced innate immunity can affect parasite density indirectly via resource depletion.     Description: Commitment to and completion of sexual development are essential for malaria parasites (protists of the genus Plasmodium) to be transmitted through mosquitoes. The molecular mechanism(s) responsible for commitment have been hitherto unknown. Here we show that PbAP2-G, a conserved member of the apicomplexan AP2 (ApiAP2) family of DNA-binding proteins, is essential for the commitment of asexually replicating forms to sexual development in Plasmodium berghei, a malaria parasite of rodents. PbAP2-G was identified from mutations in its encoding gene, PBANKA_143750, which account for the loss of sexual development frequently observed in parasites transmitted artificially by blood passage. Systematic gene deletion of conserved ApiAP2 genes in Plasmodium confirmed the role of PbAP2-G and revealed a second ApiAP2 member (PBANKA_103430, here termed PbAP2-G2) that significantly modulates but does not abolish gametocytogenesis, indicating that a cascade of ApiAP2 proteins are involved in commitment to the production and maturation of gametocytes.     Description: A functional immune system is important for survival in natural environments, where individuals are frequently exposed to parasites. Yet strong immune responses may have fitness costs if they deplete limited energetic resources or cause autoimmune disease. We have found associations between fitness and heritable self-reactive antibody responsiveness in a wild population of Soay sheep. The occurrence of self-reactive antibodies correlated with overall antibody responsiveness and was associated with reduced reproduction in adults of both sexes. However, in females, the presence of self-reactive antibodies was positively associated with adult survival during harsh winters.     Notes: A boundary element method is developed and used to compute the volume-average rheological properties of layered suspensions in a mean simple shearing flow up to a volume concentration of 0.5. The suspension is made up of a periodic array of neutrally buoyant, monosized spheroids in the x and y directions suspended in a Newtonian fluid that is subjected to a mean shear flow in the xz plane. The results for a monolayered suspension of spheres agree well with other simulations and some experimental results. Some new results for prolate and oblate spheroids are also given. Both the volume-average first and second normal stress differences are found to be zero in the simulation in all the cases considered.

    Notes: Abstract Numerical simulations of a spherical particle sedimenting in circular, triangular and square conduits containing a viscous, inertialess, Newtonian fluid were investigated using the Boundary Element Method (BEM). Settling velocities and pressure drops for spheres falling along the centre-lines of the conduits were computed for a definitive range of sphere sizes. The numerical simulations for the settling velocities showed good agreement with existing experimental data. The most accurate analytic solution for a sphere settling along the axis of a circular conduit produced results which were almost indistinguishable from the present BEM calculations. For a sphere falling along the centre-line of a square conduit, the BEM calculations for small spheres agreed well with analytic results. No analytic results for a sphere falling along the axis of a triangular conduit were available for comparison. Extrapolation of the BEM predictions for the pressure drops, to infinitely small spheres, showed remarkable agreement with analytic results. For the circular conduit, the sphere's settling velocity and angular velocity were computed as a function of drop position for small, medium and large spheres. Excellent agreement with a reflection solution was achieved for the small sphere. In addition, end effects were investigated for centre-line drops and compared where possible with available experimental data and analytic results.

    Notes: Abstract A variety of studies reported in the literature have established that initially well mixed suspensions subjected to non-homogeneous shear flows attain an anisotropic particulate structure. It has also been shown that non-homogeneous shearing causes suspensions of unimodal spheres to demix, i.e., gradients in solids concentration are formed. The objective of this study was to determine the effect of non-homogeneous shear flows on suspensions of bimodal particles, and specifically, to determine if the solids concentration gradients which develop are accompanied by size segregation of the coarse with respect to the fine fraction. We used the simplest and most direct methods to determine the relative solids concentrations: visual observation of tracer particles in transparent suspensions and physical separation of the coarse and fine solid fractions. Three different types of non-homogeneous shear flows were examined, and in each case the data support two main conclusions: 1) suspended particles migrate from regions of high shear rate to regions of low shear rate, and 2) the coarse fraction of particles migrates much faster than the fine fraction, leading to size segregation of initially well-mixed suspensions. While the former conclusion is consistent with other studies reported in the literature, to our knowledge this paper provides the first data supporting and, to a limited extent, quantifying the latter conclusion.
